The structure can be incorporated into the optical probe of a spectrophotometric instrument and includes a plurality of optical send fibers (116) having input and output ends and an optical light mixer having input and output ends. The light mixer assembly (110) effectively splices the send fibers (116) to the mixer fiber (152) and includes a connector formed by outer shell (160), inner shell (162), send fiber ferrule (164), mixer fiber ferrule (166) and alignment pin (168). The send fibers (116) extend through the connector outer shell (160) and into send fiber ferrule (164). The ends of the send fibers (116) are secured (e.g., by an optical grade adhesive) in an aperture (170) in the send fiber ferrule (164), cleaved, and polished to provide an optical-quality mating surface. Similarly, the end of the mixer fiber (152) extends through the connector inner shell (162) and into mixer fiber ferrule (166). The end of the mixer fiber (152) is located within an aperture (172) in the mixer fiber ferrule (166).

A method for operating a spectrophotometric instrument of the type for measuring the oxygenation state of hemoglobin in tissue. The method includes the use of stored hemoglobin concentration relationship data characterizing the relationship between second derivative absorbance values at a hemoglobin-absorbing wavelength and hemoglobin concentration in a tissue as a function of hemoglobin oxygenation state. Data representative of a second derivative absorbance value of tissue being analyzed is received. The hemoglobin oxygenation state of the tissue is determined as a function of the second derivative absorbance value. The hemoglobin concentration in the tissue is then determined as a function of the hemoglobin concentration relationship data, the second derivative absorbance value and the hemoglobin oxygenation state. The accuracy of the hemoglobin oxygenation state can be determined as a function of the hemoglobin concentration value.

The calibration device is adapted to receive the probe and has an optical filter for transmitting the measurement light signals but not the calibration light signal. The processor/controller is coupled to the detector and initiates a calibration procedure when the calibration light signal is not detected.

From this data characterizing data, including ischemia onset slope, ischemia onset response time, ischemia recovery slope, ischemia recovery response time or hyperemia is automatically determined. This characterizing data is related to corresponding characterizing data determined from a control population. The relationship may be displayed on a graphical display.

The boot releasably mates with and provides an interference/friction fit to the probe. The tissue-engaging surface surrounds and extends from the boot. A window (102) in the base member extends into the boot to transmit light between the probe and tissue. A layer of optically transparent material covers the window on the tissue-engaging surface. Adhesive (105) on the tissue-engaging surface of the base member releasably secures the probe to the tissue being analyzed.
