Compositions and methods comprising polynucleotides and polypeptides having dicamba decarboxylase activity are provided. Further provided are nucleic acid constructs, host cells, plants, plant cells, explants, seeds and grain having the dicamba decarboxylase sequences. Various methods of employing the dicamba decarboxylase sequences are provided. Such methods include, for example, methods for decarboxylating an auxin-analog, method for producing an auxin-analog tolerant plant, plant cell, explant or seed and methods of controlling weeds in a field containing a crop employing the plants and/or seeds disclosed herein. Methods are also provided to identify additional dicamba decarboxylase variants.

Novel proteins are provided herein, including proteins capable of catalyzing the acetylation of glyphosate and other structurally related proteins. Also provided are novel polynucleotides capable of encoding these proteins, compositions that include one or more of these novel proteins and/or polynucleotides, recombinant cells and transgenic plants comprising these novel compounds, diversification methods involving the novel compounds, and methods of using the compounds. Some of the novel methods and compounds provided herein can be used to render an organism, such as a plant, resistant to glyphosate.

Compositions and methods are provided to detoxify an auxin-analog herbicide through the use of at least one GH3 polypeptide having amino acid/auxin analog herbicide conjugation activity. Such GH3 polypeptides in the presence of an auxin-analog herbicide will produce an amino acid/auxin-analog conjugate having reduced herbicidal activity. Various methods of employing the GH3 polypeptides and the polynucleotides encoding the same are provided. Such methods include methods to detoxify an auxin-analog herbicide comprising applying to a plant, a plant cell or a seed an auxin-analog herbicide, wherein the plant, plant cell or seed comprises a heterologous polynucleotide encoding a GH3 polypeptide, and expression of the GH3 polypeptide produces a non-herbicidal aspartate/auxin-analog conjugate or a glutamate/auxin-analog conjugate. Additional methods for controlling at least one weed in an area of cultivation comprising a crop or a seed of the crop are provided, as are methods of detoxifying a contaminated material having an auxin-analog herbicide.

Methods and compositions are provided related to improved plants that are tolerant to more than one herbicide. Particularly, the invention provides plants that are tolerant of glyphosate and are tolerant to at least one ALS inhibitor, and methods of use thereof. The glyphosate/ALS inhibitor-tolerant plants comprise a polynucleotide that encodes a polypeptide that confers tolerance to glyphosate and a polynucleotide that encodes an ALS inhibitor-tolerant polypeptide. In specific embodiments, a plant of the invention expresses a GAT polypeptide and an HRA polypeptide. Methods to control weeds, improve plant yield, and increase transformation efficiencies are provided.
