Biological samples are analyzed for benzodiazepines in a single isocratic analysis using a chromatographic column system containing an immobilized enzyme reactor which cleaves glucuronic acid-conjugated benzodiazepines, an anion exchange column, a hydrophobic cation exchange column and a reverse-phase analytical column. Preferred methods of performing the analysis further involve the use of a hydrophobic cation exchange precolumn prior to the anion exchange column. The system readily lends itself to automation, automatic periodic sampling and benzodiazepine identification and quantification. The system is particularly well adapted to the determination and identification of benzodiazepines in urine samples.

A measuring part (2) measures the absorbance spectrum of an effluent fluid sample from a chromatograph in a constant cycle, and a base line calculating part (4) calculates a base line including absorbance values on both ends of a designated wave number range with respect to every absorbance spectrum data as measured. A peak area calculating part (6) calculates the peak area of the absorbance spectrum in the designated wave number range on the basis of the decided base line. A chromatogram is obtained by time change of the calculated peak area. Thus, it is possible to eliminate influences by other functional groups and fluctuation of the base line in an absorbance analyser.

This invention is a method for a substrate concentration measurement method comprising measuring a concentration of a substrate in a blood specimen containing hemoglobin. In the method ,the substrate concentration is calculated by using a measurement value correlating with a substrate concentration influenced by hemoglobin and a hemoglobin concentration or a value correlating with the hemoglobin concentration. For the substrate concentration is substrate concentration measurement method according to claim 1, wherein the substrate concentration is calculated by correcting the measurement value Re correlating with the substrate concentration based on the following Formulas 1 and 2: Corrected value = 100 × Re / V % , wherein V (%) represents a value with respect to plasma (a ratio of a measurement value influenced by hemoglobin with respect to a measurement value obtained by measuring plasma); and Re represents a measurement value correlating with a substrate concentration influenced by hemoglobin.
