摘要: PROBLEM TO BE SOLVED: To obtain a new gene involved in the differentiation maintenance of smooth muscle cells, to obtain a protein encoded by the gene, to provide a method for diagnosing diseases caused by the abnormal proliferation of smooth muscle cells with the gene's expression as the indicator, and to obtain a diagnostic for the diseases. SOLUTION: In culturing chicken gizzard smooth muscle cells, an insulin-like growth factor(IGF-I) is added to develop a culture system suitable for maintaining the phenotype of smooth muscle cell-differentiated type; using this culture system, a macroarray analysis is carried out for 681 clone screened by differential display method and 2016 clone made by using subtracted hybridization method, and 79 new genes (EST clones) of smooth muscle cell- differentiated type with raised expression are isolated; further, it is found that 13 known genes are involved in the differentiation maintenance of smooth muscle cells; by detecting the expression of these genes, diagnosis of diseases including arteriosclerosis, myocardial infarction, aortic aneurysm, cerebral apoplexy and glomerulonephritis, for each of which the phenotype transformation of smooth muscle cells is considered as a cause, is expected.
