Abstract: Pol γ protein inhibitor compounds are useful for selectively inhibiting the growth of MLH1 mutant/deficient tumor or cancer cells due to a synthetic lethal relationship between MLH1 and Pol γ. The compounds were shown to directly interact with Pol γ and inhibit its biological function and cause cancer cell death and an inhibition of tumor or cancer cell growth in multiple assays.
