摘要: The present application refers to the development of a new effective antidote for the poisonous Amanita mushroom species, after exploring an innovative in silico and in vivo approach based on the binding site of amatoxin to RNA polymerase II (RNAPII) and the screening of clinical drugs with bioisosterism with amatoxins in the same models. Proof of concept was attained in vivo, using CD-1 mice, and clinical application is immediately proposed, in addition to the already prescribed therapeutic measures, taking advantage of well-established clinical use of the drug found to be an effective antidote, polymyxin B. Thus, Polymyxin and/or its derivatives/precursors consist in a therapeutic strategy on Amanita Phalloides as demonstrated by data gathered and showed in the present application.
