Aspects of the disclosure relate to compositions and methods useful for treating Huntington' s disease. In some embodiments, the disclosure provides interfering nucleic acids (e.g., artificial miRNAs) targeting the huntingtin gene (HTT) and methods of treating Huntington's disease using the same. Accordingly, in some aspects, the disclosure provides an isolated nucleic acid comprising or encoding the sequence set forth in any one of SEQ ID NOs: 1-22.

A system includes a probe assembly, a camera, and a control system. The probe assembly includes a rigid substrate, a compliant layer provided on the rigid substrate, one or more rigid probes arranged on the compliant layer to cover at least a portion of the compliant layer, and a reflective layer covering the one or more rigid tips and uncovered portions of the compliant layer. The camera is configured to generate image data from the probe assembly. The control system is configured to receive image data from the camera and develop a topographical image of a surface of a sample, based at least in part on the received image data.

A method for obtaining one or more images of a sample using a microscope includes dividing, using a reverberation cavity, a first one of a plurality of laser pulses into a plurality of sequential sub-pulses, each of the plurality of sequential sub-pulses having a power that is less than a previous one of the plurality of sequential sub-pulses, directing, using the one or more lenses of the microscope, the plurality of sequential sub-pulses onto a portion of the sample to generate a plurality of signals, each of the plurality of signals being associated with a different depth within the sample, and detecting the plurality of signals from the sample to generate one or more images of at least a portion of the sample.

A system for analyzing a tissue sample includes two wavelength-division multiplexers and a fiber coupler. The first wavelength-division multiplexer combines visible and near infrared electromagnetic radiation and directs the combined electromagnetic radiation to the fiber coupler. The fiber coupler emits a sample beam of visible and near infrared electromagnetic radiation toward a tissue sample, and a reference beam of visible and near infrared electromagnetic radiation toward a reference mirror. The sample beam reflects off the tissue sample back to the fiber coupler. The reference beam reflects off the reference mirror back to the fiber coupler. The fiber coupler combines the reflected sample and reference beams and directs the combined electromagnetic radiation to the second wavelength-division multiplexer. The second wavelength-division multiplexer sends visible electromagnetic radiation from the sample and reference beams to a first spectrometer, and near infrared electromagnetic radiation from the sample and reference beams to a second spectrometer.

As demonstrated herein, a preferential and progressive expansion of a subset of hematopoietic cells bearing somatic mutations in one or more HSC cardiometabolic driver genes leads to pro-inflammatory signaling at multiple levels. Accordingly, provided herein are compositions, methods, and assays for modulating a HSC cardiometabolic driver gene mutation-mediated proinflammatory activity.
Abstract:
There are gluten-degrading enzymes found in Rothia species bacteria that are subtilisins that belonging to the S8A family of serine protease family. The Rothia sp. derived subtilisin-like enzymes have the conserved catalytic triad composed of a Ser, His, and Asp residues that is characteristic of the serine protease family. The Rothia subtilisin enzymes are potent at cleaving proline-containing proteins, cleaving the second peptide bond after proline in the XPX1 motif, where X is any amino acid, P is proline and X1 is a hydrophobic amino acid, e.g. the XPQ motif, where Q is glutamine. Embodiments herein provide isolated enzyme compositions and formulations comprising subtilisins gluten-degrading enzyme from a Rothia species bacteria. Also provided herein are methods of treatment of celiac disease or a related disorder, treatment of gluten-containing foodstuff, degrading and/or detoxifying gluten comprising the subtilisins gluten-degrading enzyme and/or compositions.
