The emergence of a new betacoronavirus SARS-CoV-2 has led to a major health-related crisis associated with a significant mortality in intensive care units, due to the pulmonary complications of COVID-19. The inventors showed that BAY-2402234 is suitable for inhibiting replication of coronavirus and thus would be suitable for the treatment of infections mediated by said type of virus.

The inventors surprisingly found that FXR plays a determinant role in the maintenance of viral episome in cells from tissues that are not specialized in bile salt synthesis and transport as the liver or the intestine. In particular, the inventors show that FXR agonist could be suitable for inhibiting the replication of viruses (e.g. BKV and HIV-1) that persist in the cell in an episomal and extrachromosomal form of DNA. Accordingly the present invention relates to a method of reducing persistence and expression of an episomal virus in a subject in need thereof comprising administrating to the subject a therapeutically effective amount of a FXR agonist.

L'invention concerne l'utilisation de bactéries lactiques pour traiter les maladies métaboliques, notamment le diabète de type 2, ainsi qu'une méthode de criblage de bactéries susceptibles d'améliorer la santé métabolique et permettant de diminuer, ou ralentir, la progression des maladies métaboliques. Par l'utilisation de modèles de drosophiles et de souris, les inventeurs ont démontré les effets bénéfiques de certaines souches de bactéries sur le métabolisme via la mesure de plusieurs paramètres, notamment le test oral au glucose. Plus particulièrement, l'invention concerne une méthode de criblage de bactéries à tropisme intestinal chez un mammifère dans lequel on monocolonise des larves de drosophiles et/ou des souris initialement axéniques par la souche de bactérie à tester, et on élève ces larves avec un milieu nutritionnel hypercalorique, ce qui permet de déterminer un impact favorable de la bactérie dans le cadre d'un régime hypercalorique et d'inférer un effet positif sur la santé métabolique.

The emergence of a new betacoronavirus SARS-CoV-2 has led to a major health-related crisis associated with a significant mortality in intensive care units, due to the pulmonary complications of COVID-19. The main aim of the inventors is to explore the respective efficacy of IFN-alpha polypeptides in controlling SARS-CoV-2 replication and pathogenesis of coronavirus disease 19 (COVID-19). They have therefore tested the anti SARS CoV-2 efficacy of different IFN-alpha subtypes. In particular, SARS CoV-2 susceptible target cells (Vero E6) were pretreated with each of the subtypes of IFN-alpha, or the single IFN-beta subtype. As a positive control of inhibition in this system; Remdesivir was used. 24h post-treatment, cells were infected with a strain of SARS CoV-2 for 2h, cells were then washed and IFNs were added again to the culture medium. 54h later, virus present in the supernatant was quantified by RNA extraction and RT-qPCR performed on the viral NS14 gene. They showed that some IFN-alpha polypeptides, in particular IFN-alpha 8, 10 or 14 polypeptides are particular suitable for inhibiting the replication of the virus.

The present invention relates to a non-invasive, specific and rapid diagnostic method of Familial Mediterranean fever (FMF) in a subject said method comprising the step of measuring the level of cytokine (IL-18 or IL-1 beta) secreted by immune primary cells (or cell death level of these cells) obtained from said subject, which have been beforehand treated with a Protein Kinase C (PKC) inhibitor, and optionally beforehand treated with a NF-kB activator such as LPS. Inventors show based on the extensive study of the inflammasome process of the monocytes, that PKC superfamily inhibitors trigger inflammasome activation in monocytes from FMF patients while they are not sufficient to do so in monocytes from healthy donors (HD) or from patient having hyperimmunoglobulinemia D syndrome (HIDS). Using cytokine release quantification or determination of real time cell death kinetics, inventors demonstrate that PKC superfamily inhibitors can discriminate FMF patients from HD or from patients with systemic inflammation from other aetiologies. These results thus set-up the basis for the development of a rapid functional specific diagnostic test for FMF. Methods of treatment are disclosed.
