S. pyogenes nucleic acid molecules encoding polC, dnaE, holA, holB, dnaX, dnaN, SSB, dnaG, dnaQ, dnaA and dnaB proteins, as well as nucleic acid molecules comprising oriC are provided. The encoded proteins are also provided. The nucleic acid molecules and proteins are useful for reconstituting replicases and polymerases for sequencing, amplification, and screening for compounds which modulate the function of the polymersase or replicase.

The present invention provides a method for isolating protein complexes from a cell, a cell or tissue lysate or an whole organism by employing a combined set of affinity tags of high affinity, specificity and ease of elution. The method involves using a known protein modified to contain more than one affinity tag separated by one or more specific protease cleavage sites, as bait, to isolate any interacting proteins or fragments thereof. The proteins or fragments thereof contained in the isolated complex can then be identified and the interacting partners can be used as new targets for diagnostic tools or the basis for the development of new compounds for therapeutic drug intervention.

The invention features bone grafts (such as bone allografts) with reduced protease activity. These bone grafts are useful, for example, in conjunction with a polypeptide of interest (such as platelet derived growth factor) for treating, stabilizing, preventing, and/or delaying a bone, periodontium, ligament, cartilage, or tendon condition in an individual (such as a human). Additionally, the invention provides methods of measuring the protease activity associated with a bone graft, reducing the level of protease activity associated with a bone graft, selecting a bone graft with an acceptable level of protease activity, and methods of administering a bone graft and a polypeptide of interest to an individual.
