    Abstract: Intradermal (ID) vaccination aims at targeting the large network of skin antigen-presenting dendritic cells (DC) and constitutes a promising approach to improve anti-infectious immunity. Yet, its anti-viral protective efficacy for sub-unit vaccines and the underlying skin innate immune mechanisms remains to be explored for a wider usage in humans. Using a pig model of lethal respiratory infection with pseudorabies virus (PRV), we show that ID vaccination with adjuvant-free PRV glycoproteins triggers cellular and humoral immunity and confers protection from disease induction by nasal viral challenge, as efficiently as an oil- adjuvanted reference vaccine delivered intramuscularly.
