The present Invention provides block copolymers with improved thermogelling and rheological properties due to their specific structural composition. These block copolymers are characterized by one of the general chemical structures [A] n -[B] m or [B] n -[A] m , wherein [A] is a poly(2-oxazine), and wherein [B] is a poly(2-oxazoline). The present invention additionally provides methods of synthesis as well as uses for such block copolymers.

A composition for use in treating a condition associated with degeneration of articular cartilage and/or with subchondral bone loss is disclosed herein. The composition comprises a conjugate which comprises a polypeptide having attached thereto at least two polymeric moieties, at least one of the polymeric moieties exhibiting a reverse thermal gelation. Further disclosed is a composition comprising the aforementioned conjugate along with a hyaluronic acid, an anti-inflammatory agent, an analgesic, a growth factor, a blood fraction, a nucleic acid, and/or a cell, the composition being an aqueous composition which forms a hydrogel at a temperature in a range of from 32 °C to 37 °C, as well as a method utilizing such a composition comprising a nucleic acid for effecting gene delivery. In an embodiment, the composition comprises a conjugate of fibrinogen to F127 poloxamer diacrylate.

The induction of PD-L1 and reduction of HLA-DR by TLT-1 or TLT-1-derived polypeptides can therefore be useful in treating diseases associated with immune hyper-reactivity, such as autoimmune diseases, hypersensitivity reactions, transplantation rejections and graft versus host disorders.

In the present disclosure, polymer conjugates, including polymer-antibody-drug conjugates (polymer ADCs) are described, as well as the use of such conjugates to treat human disease. The polymer conjugates can contain a large number of polymer-bound agents, thus effectively increasing the drug antibody ration (DAR) of the antibody significantly beyond the currently available technology. This may be of particular importance when antibodies to low density antigens are used as target antibodies. The described polymer-ADCs have improved pharmacokinetics and solubility relative to traditional ADCs. The linker between agent and the polymer can be tailored to provide release of toxin at the desired site and under the desired conditions within the tumor. An additional feature of the polymer-ADCs of the current disclosure is that a purification moiety can be attached to the polymer backbone to provide ease of purification of the polymer-ADCs.
