Abstract: Compositions and methods are provided for identifying and measuring biomarkers in hair. In particular, methods for extracting proteins and RNA species from hair, and for identifying and measuring hair biomarkers are disclosed. Serial segmentation of a hair sample can be used to assess sequential epochs of protein expression, metabolic regulation, and gene expression in the body across different time periods. Use of hair biomarkers provides a non-invasive, pain-free method for diagnosing diseases and identifying individuals at risk for poor health outcomes.

Abstract: Disclosed are methods, libraries, and samples for quantifying a target analyte in a laboratory sample including the target analyte. The methods typically include the step of estimating the amount of the target analyte in the laboratory sample from mass spectrometric data including signal intensities for the target analyte and one or more internal standards, where the mass spectrometric data are an output of a mass spectrometric analysis of a target sample produced from the laboratory sample and a predetermined amount of the one or more internal standards. The present disclosure also provides a method for analyte quantification. The method comprises adding one or more calibrators to a sample comprising one or more analytes; applying mass spectrometry (MS) to the sample; and using a trained machine learning model to determine an absolute concentration of the one or more analytes.

Abstract: Disclosed herein are buffer solutions for decreasing the size of particles during imaging. The buffer solutions may comprise polymers, ions, or a combination thereof. The buffer solutions may decrease the sizes of particles. Decreasing the sizes of the particles during imaging can improve the resolution of individual particle imaged, and may reduce particle aggregation and/or increase substrate loading efficiency. In some cases, decreasing the sizes of the particles can improve the resolution of individual reads of sequencing. Also provided herein are methods and systems of using the same.

Abstract: An apparatus and method for removing bubbles from a fluid sample. The apparatus has a barrel and a filter member. The filter member is slidably disposed within the internal chamber and defines an inlet side and an outlet side of the internal chamber. The filter member has a gas-permeable, liquid-impermeable membrane which permits the gas portion of the fluid sample to pass across the filter member. The barrel has a second gas-permeable, liquid-impermeable membrane which provides a fluid-tight seal across the outlet opening.

Abstract: Analyzing a buried layer on a sample includes milling a spot on the sample using a charged particle beam of a focused ion beam (FIB) column to expose the buried layer along a sidewall of the spot. From a first perspective a first distance is measured between a first point on the sidewall corresponding to an upper surface of the buried layer and a second point on the sidewall corresponding to a lower surface of the buried layer. From a second perspective a second distance is measured between the first point on the sidewall corresponding to the upper surface of the buried layer and the second point on the sidewall corresponding to the lower surface of the buried layer. A thickness of the buried layer is determined using the first distance and the second distance.
