    摘要: The present invention relates to a binding motif of a receptor. In particular, the binding motif is a cytoplasmic domain associated with stimulation of receptor mediated activities. The present invention further contemplates methods of using the motif in particular for mediating activities of receptors. In one aspect of the present invention, there is provided a binding motif of a receptor capable of binding a cytoplasmic protein, said binding motif comprising an amino acid sequence wherein at least one amino acid is serine/threonine preferably there are at least two serines to indicate the cytoplasmic binding motif. Preferably the binding motif comprises an amino acid sequence including the sequence: -S-X-S/T- wherein the X is any amino acid.

    摘要: A therapeutic agent capable of binding to the F'-G' loop of domain 4 of the common β c chain of the receptor for GM-CSF, IL-3 and IL-5. The amino acid Tyr 421 which is located in the F'-G loop is critical in the high affinity binding and stimulation of function of GM-CSF, IL-3 and IL-5 to the common β c chain of their receptors. Other receptors for cytokines also show hydrophobic amino acids in analogous positions and it is probable that they too will play a pivotal role in binding of respective cytokines to them and in modulating function. Agents capable of binding to the F'-G' loops are suggested and should be of therapeutic value.

    摘要: A method of isolating a monoclonal antibody capable of inhibiting any one of IL-3, GM-CSF and IL-5 binding to the common receptor beta c, or a monoclonal antibody capable of inhibiting the cytokines binding to a receptor analogous to beta c. The method includes the steps of immunising an animal with a cytokine receptor or portion of a cytokine containing the critical binding site which portion includes the extracellular domain 4 or analogous domain in the analogous common receptor or part thereof. Antibodies producing cells from the animal are then isolated and fused with a myeloma cell line and then screened for a cell line that produces an antibody of the desired type. A monoclonal antibody, or fragments thereof capable of inhibiting the binding of the cytokines IL-3, GM-CSF and IL-5 to the beta c receptor, and a hybridoma cell line producing the antibody are also claimed.
