The present invention relates to anti-thrombin polypeptides isolated from the leech Hirudinaria manillensis and to processes for their preparation. The polypeptides of the invention may be modified by way of amino acid extension at either or each end, and may be subjected to post-translational modification. The anti-thrombin polypeptides may be prepared by isolating them from the tissue or secretions of the leech Hirudinaria manillensis but they also may be synthetized by recombinant DNA methods. According to this latter aspect, the invention provides DNA sequences, expression vectors and host cell lines for the recombinant preparation of the polypeptides. The anti-thrombin polypeptides of the invention find an useful application in the treatment of venous thrombosis, vascular shunt occlusion and thrombin-induced disseminated intravascular coagulation.

A gene coded for a polypeptide isolated from an occlusion body of a baculovirus such as Trichoplusia ni granulosis virus possessing a biological activity of enhancing virus infection of host insects by causing rapid degradation of the peritrophic membrane lining the midgut lumen of insects. A polyhedrin gene of Spodoptera litura nuclear polyhedrosis virus (SℓNPV C-411) has the following restriction enzyme cleavage map and about 3 kilobase pairs: At least a part of said polyhedrin gene can be substituted by a gene coding for a useful substance to construct a vector and a recombinant virus, and it is possible to produce a useful substance such as a peptide, protein or glycoprotein by culturing a cell infected with the recombinant virus.

A method for producing a recombinant baculovirus expression vector, capable of expressing a selected gene in a host insect cell is disclosed. The method involves cleaving baculovirus DNA to produce a DNA fragment comprising a polyhedrin gene or portion thereof, including a polyhedrin promoter. A recombinant transfer vector is prepared by inserting said DNA fragment into a cloning vehicle and thereafter inserting a selected gene into the thus modified cloning vehicle such that it is under the transcriptional control of the polyhedrin promoter. The recombinant transfer fector is contacted with a baculovirus DNA so as to effect recombination and incorporation of the selected gene into the baculovirus genome. The resultant recombinant baculovirus is then used to infect susceptible insects or cultured insect cells and the protein product from the incorporated selected gene is produced from the infection.
