The present invention may be included in the field of biotechnology and it covers the improved production of recombinant proteins in insect cells or insect larvae as biofactories by a novel expression cassette. This expression cassette comprises nucleic acid sequences such as promoters, homologous regions ( hr ) as enhancers, and sequences encoding transcriptional regulators, for example, the baculovirus Ac-ie-01 cDNA, or any combination thereof, which are able to increase the quality and production efficiency of the recombinant proteins. Moreover, the present invention is also directed to the vectors themselves comprising the above mentioned nucleic acid sequences of the invention, cells or insects infected, transformed or transfected with those sequences or vectors, and methods for producing the recombinant proteins by using the aforesaid sequences, vectors, cells or insects.

In this method, the desired protein is produced in the form a fusion protein with a protein constituting a virus particle, and the virus particle is recovered. Nuclear polyhedrosis viruses, for example, Autographa californica nuclear polyhedrosis virus (AcMNPV), useful in the control of lepidopterous larvae such as the larvae of the cabbage looper Trichoplusia ni , have been found to have enhanced infectivity when mixed with certain proteins obtained from the granulin fraction of Trichoplusia ni granulosis virus (TnGV) or Heliothis armigera granulosis virus (HaGV), and from the polyhedrin fraction of AcMNPV viruses. The proteins from the TnGV granulin fraction have molecular weights between 101 and 104 Kd. The enhanced infectivity is correlated to biochemical and structural changes in the T.ni peritrophic membrane.

A gene coded for a polypeptide isolated from an occlusion body of a baculovirus such as Trichoplusia ni granulosis virus possessing a biological activity of enhancing virus infection of host insects by causing rapid degradation of the peritrophic membrane lining the midgut lumen of insects. Novel pesticides incorporating said gene or fragments thereof are also claimed.

Recombinant Bombyx mon nuclear polyhedrosis viruses having, in the polyhedral protein-encoding structural gene portion of the Bombyx mon nuclear polyhedrosis virus DNA (BmNPV DNA), a structural gene for a desired protein as joined to the whole of or a part of the polyhedral protein-encoding structural gene with or without interposition of a linker base sequence are provided. By propagating the above viruses in silkworm-derived cells or silkworms, fused proteins of a desired protein and the whole of or a part of the polyhedral protein as joined together with or without interposition of a linking amino acid or peptide are produced. The desired proteins can be obtained from the fused proteins by cleavage or decomposition at the linking site.

A recombinant baculovirus displaying on its envelop a fusion protein is disclosed. The fusion protein comprises a heterologous antigen, and a C-terminal region of baculovirus envelope GP64 protein, which has at least 100 amino acid residues in length and lacks a B12D5 binding epitope located within the central basic region of the GP64 protein. The genome of the recombinant baculovirus comprises a transgene encoding a fusion protein that comprises a signal peptide, the heterologous antigen, and the C-terminal region of the baculovirus envelope GP64 protein, in which the antigen is located between the signal peptide and the C-terminal region of the GP64 protein. Methods for eliciting an antigen-specific immunogenic response in a subject in need thereof are also disclosed.

The present invention provides a novel transfer vector and a recombinant baculovirus; methods for the production thereof; and pharmaceuticals containing the recombinant baculovirus as an active ingredient, which are useful as preventive or therapeutic drugs for infectious diseases such as malaria and influenza. More specifically, the invention provides a recombinant transfer vector capable of expressing a foreign gene fused to a virus gene under the control of a dual promoter; a recombinant baculovirus; methods for the production thereof; and pharmaceuticals containing the recombinant baculovirus as an active ingredient.
