The present invention relates to a method for the cultivation of primary cells. The primary cells are cultivated in a serum free medium comprising a factor selected from the group consisting of growth factors and attachment factors. The method for the cultivation of primary cells may be one step in a method for the amplification of viruses, such as poxviruses. According to this latter method the primary cells are cultivated in a serum free medium comprising a factor selected from the group consisting of growth factors and attachment factors. The cells are then infected with the virus and the infected cells are cultivated in serum free medium until progeny virus is produced.

Compositions and methods are described for generating an improved effective immune response against an immunogen in humans. The enhanced immune response, is obtained by using an MVA vector as a prime and an adenovirus vector as a boost and is characterized by a high level of antibody response specific to the immunogen, and an enhanced cellular immune response. The compositions and methods can be used to provide a protective immunity against a disease, such as an infection of one or more subtypes of Ebola and Marburg filoviruses, in humans.

The invention is drawn to compositions and methods for the induction of a strong CD8 T cell response to a specific antigen(s). The combination of an early/late hybrid promoter directing strongly enhanced early expression of a neoantigen with at least three immunization rounds resulted in a highly efficient neoantigen-specific CD8 T cell response. This combination reversed the immunodominance hierarchy and converted a moderately immunogenic and subdominant CD8 T cell epitope into the immunodominant epitope.
