The method applies a Top Hat algorithm to the image then constructs inner and outer markers for application to the original image in a Watershed algorithm. The inner marker is constructed using binary erosion. The outer marker is constructed using binary dilation and perimeterisation. The method finds particular application for first level segmentation of a cell nucleus prior to detailed analysis.

A method of segmentation of cell nuclei is described that uses an active contours approach based on a Viterbi search algorithm. Also disclosed is an initialisation technique that maximises the probability of a correct segmentation. The invention also includes a method to classify cell nuclei according to the difficulty of correct segmentation. Nuclei that are difficult to segment can be rejected to minimise the probability of false positives. The probability of false positives can be driven close to 0 %.

These time series are regarded as samples from some underlying distribution, and statistical techniques are used to identify the density function of this distribution. The background can then be identified as a feature of this distribution, moreover the distribution of features is subsequent image frames can be monitored to see of it deviates sufficiently from the current distributions to indicate a change in the background.

Each binary representation in the first set of binary representations is mapped to a further set of binary representations using a second monotonically varying operator, such as a spatial operator. The result of the two mappings is a matrix of binary image representations. Each binary image representation is allocated a scalar value to form an array of scalar values which may be analysed to identify defined texture characteristics.

A method of segmenting chromatin particles in a nucleus of a cell by locating regional minima in an image, computing a zone of influence (ZOI) around each regional minimum, and segmenting a single chromatin blob within each ZOI using a region growing procedure. The method can be used as the basis of a method of qualitatively characterizing the distribution of nuclear chromatin by computing features for individual chromatin particles. Chromatin features can be synthesized from the features of individual particles and particle features can be synthesized into nucleus features and slide features. The method is useful for detecting malignancy associated changes and changes during neoplasia. The method may also be used more generally to assess chromatin patterns in living cells during the cell life cycle. This makes it possible to measure alternations in the evolving patterns that result from pathological or environmental influences.
