This invention is based on the unexpected discovery that transgenic mice containing a nucleic acid construct encoding the SV40 early region tsA58 mutant regulated by the rat PF4 promoter unexpectedly develop varying levels of thrombocytopenia and/or megakaryocyte leukemia. In accordance with this invention, the mice present a novel and useful system for screening compounds useful for treating and/or preventing these diseases.

Disclosed are novel proteins, referred to as megakaryocyte growth and development factors (MGDFs; also generally referred to as Mpl ligands), that have a biological activity of stimulating the growth of megakaryocytes and augmenting the differentiation or maturation of megakaryocytes, ultimately to result in the production of platelets. MGDF derivatives comprising MGDF molecules attached to water soluble polymers, such as polyethylene glycol, are also disclosed, along with methods for their preparation. Also disclosed are processes for obtaining the MGDFs in homogeneous form from natural sources and producing them by recombinant genetic engineering techniques from mammals, including humans.

Non-naturally occurring polypeptide (I) is claimed, whose aminoacid sequence is similar enough to naturally-occurring stem cell factor (SCF) to give it the same haematopoietic biological activity. (I) is produced by eukaryotic or prokaryotic expression of an exogeneous DNA sequence, and has in vivo and in vitro biological activity. Also claimed are a DNA sequence encoding (I), a host transformed with the DNA sequence, a process for producing and recoverng SCF, a DNA sequence encoding a human SCF analogue, an antibody that binds SCF, a compsn.; of (I) with a water-soluble polymer, a method of preparing the above compsn. and a method of transfecting early haematopoietic stem cells.
