    Abstract: PROBLEM TO BE SOLVED: To provide a novel molecule for antisense use, hardly degraded by nuclease in vivo, having high binding affinity and specificity to target mRNA, and capable of efficiently controlling expression of a specific gene.SOLUTION: An novel artificial nucleoside of the invention has an amide bond introduced into a crosslinked structure of 2',4'-BNA/LNA. An oligonucleotide comprising the 2',4'-crosslinked artificial nucleotide has binding affinity to single stranded RNA equivalent to known 2',4'-BNA/LNA, and nuclease resistance exceeding LNA. In particular, due to its much stronger binding affinity to single stranded RNA than S-oligo, its application to nucleic acid medicine is expected.
