PROBLEM TO BE SOLVED: To provide a bacterium belonging to the genus Trichoderma capable of controlling white root disease in Hevea brasiliensis, an agent for controlling a plant pathogen and a soil improver using the same, and a method for controlling a plant pathogen and a method for improving soil.SOLUTION: A bacterium belonging to the genus Trichoderma is antagonistic to the bacterium that causes white root disease in Hevea brasiliensis. An agent for controlling a plant pathogen includes the cultured product, wet biomass, or dry biomass of the bacterium belonging to the genus Trichoderma. A soil improver includes the cultured product, wet biomass, or dry biomass of the bacterium belonging to the genus Trichoderma. A method for controlling a plant pathogen includes a step of treating soil using the agent for controlling a plant pathogen. A method for improving soil includes a step of treating soil using the soil improver.

PROBLEM TO BE SOLVED: To provide a method for specifically detecting Rigidoporus microporus of Hevea brasiliensis white root disease microbe, living in soil and plants in high sensitivity in order to detect Hevea brasiliensis white root disease in an early stage.SOLUTION: This method for detecting Hevea brasiliensis white root disease microbe carries out a PCR by using a forward primer specifically hybridizing with a specific region in an ITS region of rDNA of Rigidoporus microporus, and a reverse primer specifically hybridizing with a specific region in an ITS region of rDNA of Rigidoporus microporus, by bringing a DNA extracted from a microorganism-containing sample as a template.

According to the present invention, a gene cluster encoding various kinds of prenyl transferase of Hevea brasiliensis is obtained, and the nucleotide sequences of these genes are determined. The prenyl transferases are involved in the biosynthesis of isoprene backbone of natural rubber. Therefore, improvement of productivity of rubber can be expected, by transforming plants by the gene cluster obtained in the present invention.

PROBLEM TO BE SOLVED: To provide a method for isolating a large quantity of live laticifer cells from Hevea brasiliensis by one time operation.SOLUTION: This method for isolating laticifer cells comprises (a) a step of obtaining an enzyme digested solution by preparing an enzyme treated solution in which a sample of Hevea brasiliensis is brought to contact with an enzyme solution to treat the sample with the enzyme, (b) a step of obtaining a supernatant by centrifuging the enzyme digested solution obtained in the step (a) and (c) a step of obtaining a laticifer cell suspension from the supernatant by filtering the supernatant obtained in the step (b).
