PROBLEM TO BE SOLVED: To provide a mutant AD4 protein related to Alzheimer's disease and a gene encoding the protein. SOLUTION: The mutant AD4 protein has a specific amino acid sequence (provided that the amino acid residue 141 is isoleucine). The protein can be separated as a recombination translation product by culturing a proper host and vector system. The supernatant derived from such cell strain, an enclosed protein when the protein is not excreted in the supernatant, or the whole cell can be treated by various refining procedures for the separation of the desired protein. PROBLEM TO BE SOLVED: To provide new thymidine kinase mutants and TK (thymidine kinase) fusion proteins with properly increased biological activity for various applications (for example, gene therapy), and other related merits. SOLUTION: The present invention provides isolated nucleic acid molecules encoding a Herpesviridae thymidine kinase enzyme comprising one or more mutations, at least one of the mutations encoding an amino acid substitution located toward the N-terminus from a DRH nucleoside binding site which increases a biological activity of the thymidine kinase, as compared to unmutated thymidine kinase. Such mutations include amino acid substitutions within a Q substrate binding domain which increases biological activity of the thymidine kinase, as compared to unmutated thymidine kinase. PROBLEM TO BE SOLVED: To provide a method and composition useful for diagnosis of patients of scurfy-associated diseases. SOLUTION: The problem is solved by providing isolated nucleic acid molecules which encode Fkh sf , as well as mutant forms thereof. Also provided are expression vectors suitable for expressing such nucleic acid molecules, and host cells containing such expression vectors. Also provided are pharmaceutical compounds and methods of identifying such compounds that can modulate the immune system. SOLUTION: The invention provides an antibody specifically bonding to the following mutant AD4 protein (a) or (b): (a) a protein composed of 448 amino acid residues and produced as a genetic product of human AD4 gene wherein the amino acid residue 141 is isoleucine; and (b) a protein composed of an amino acid sequence obtained by the deletion, substitution or addition of one or more amino acids in the protein (a) provided that the isoleucine at the 141st original amino acid sequence site is not deleted by the above deletion, and comprising a causal protein of Alzheimer's disease.
