SOLUTION: There are provided specifically a method for increasing production of highly unsaturated fatty acids by marine microorganisms of genus Crypthecodinium while growing in low chloride media by manipulating sodium ion and potassium ion levels, and a method for producing the highly unsaturated fatty acid by marine organisms at low pH levels, including a method for generating low pH tolerant strains. PROBLEM TO BE SOLVED: To provide a method for producing dinoflagellate biomass containing single-cell oil for simple fare. SOLUTION: This method for producing the dinoflagellate biomass comprises (a) adding C.cohnii of about 10 6 cell/ml (0.5-1.0 g dry weight/L) to a fermentor containing in the initial stage nutrient solutions containing about artificial seawater of quarter or half salinity, 1-8% glucose, and 0.4-0.8% yeast extract, (b) culturing C.cohnii at 15°C-34°C at 5.0-9.0 pH, (c) gradually adding glucose and yeast extract to the nutrient solutions for about 56 h, (d) further adding glucose to the nutrient solutions for about 16 h so as to induce C.cohnii to produce oil, (e) stirring and ventilating the fermentor so as to keep dissolved oxygen content to at least about 20% of aeration level through culturing, and (f) collecting C.cohnii after about 60 h to 90 h.
