PROBLEM TO BE SOLVED: To provide an active component capable of stimulating the direct or indirect expression of type 2 and/or type 3 human β-defensin without inducing inflammation, irritation or hypersensitive reaction. SOLUTION: The increase of the amount of β-defensin in human skin prevents the pathogenic specific species (Staphylococcus aureus) from invading excessively into an ecosystem of a body, and contributes for protecting the skin and keeping it to be healthy. The active component is selected from the roots of mugwort, horseweed, bark of a red-berried elder, etc., or their extract, α-MSH, one of its constituting peptides and substance resembling to the peptides. The active components can be used as a cosmetic composition containing the same by 0.01-10 wt. %. Also, the active components can be used as medicinally permissible excipients. SOLUTION: The method for testing the enzyme activity of phospholipase A2 comprises (a) and (b): (a) the material which may be effective for inhibiting the enzyme activity of phospholipase A2, preferably I type or II type of phospholipase A2 is put in the presence of a substrate which is a phospholipid containing at least one species of ester type fatty acid, preferably the fatty acid having a 15-22C long chain, more preferably an unsaturated or polyunsaturated fatty acid, and releases at least one kind of the fatty acids when it is hydrolyzed; (b) the presence of the fatty acid is detected and the quantity of the same is detected as the case may be. SOLUTION: The hydrated lamellar phase or liposome contains either a polyethylenimine, or a substance which stimulates the intracellular penetration and selected from the group consisting of a fatty monoamine of a 10-18C carbon-containing chain length and a cationic polymer, optionally at least one fluorescent compound which is essentially inert with respect to the intracellular penetration, which enables this penetration to be visualised. The cosmetic composition and the pharmaceutical composition use the hydrated lamellar phase or liposome. A substance held in the hydrated lamellar phase or in the liposome, or a substance potentially having the ability for stimulating the intercellular penetration of an active ingredient is screened by detecting the intercellular penetration of at least the fluorescent compound by measuring the intercellular fluorescence by using the hydrated lamellar phase or the liposome. SOLUTION: The active ingredient is a natural extract having the above-mentioned activity. The natural extract is a plant-derived extract such as soybean extract or oat(Avena sativa) extract or an animal-derived extract such as silk extract, fish roe extract or fish meal extract. This active ingredient can be used for producing a composition that increases active-type TGFb1 concentrations in the skin( especially corium ).
