Pharmaceutical hydrogel formulations containing polyvinyl alcohol are provided. The formulations may serve as rug reservoirs in electrotransport drug delivery systems or passive transdermal systems, or they may be used in a variety of other types of dosage forms, e.g., capsules, suppositories, aerosols, and the like. With these formulations, there is virtually no syneresis encountered upon long term storage, an advantage that derives from selecting the quantity of polyvinyl alcohol in the gel to correspond to the polymer's degree of hydrolysis.

An electrode assembly and a method of forming an anhydrous reservoir layer of an electrode assembly in an electrotransport transdermal agent delivery device are provided. The reservoir layer is adapted to be placed in agent-transmitting relation with a body surface and an electrode in electrical contact with a power source and the reservoir layer. The method includes the steps of dissolving a beneficial agent in a solvent, applying the solvent and dissolved beneficial agent to a surface of a hydrophilic polymer filtration membrane, removing the solvent from the surface of the filtration membrane, and disposing the beneficial agent/filtration membrane within the electrode assembly.

Improved electrotransport is obtained by treating the therapeutic agent with a multivalent metal compound of the formula MX where M is a metallic cation having a valency of at least +2 and is reactive with the polybasic anionic counter ion and X is a pH-increasing anion. Reduction in species which compete with the therapeutic agent for electrotranport is obtained.

A method is provided for preparing a novel therapeutic agent-containing reservoir (26, 28) for use in conjunction with an electrotransport drug delivery system (10). A polymeric matrix is foamed in a selected atmosphere and then cross-linked to form a polymeric closed-cell foam matrix reservoir (26, 28). The method enables relatively smaller quantities of therapeutic agent to be loaded into the electrotransport system (10) and is especially useful for the transdermal delivery of more costly drugs, such as peptides and proteins produced from genetically engineered cell lines, and/or highly potent drugs for which a small dosage is efficacious. Reservoirs (26, 28) prepared according to this method and electrotransport devices (10) containing such reservoirs are also provided.

A transdermal electrotransport drug delivery device having an anode, a cathode and a source of electrical power electrically connected to the anode and the cathode. The cathode includes a cathodic electrode and a cathodic reservoir comprised of a housing composed of a polymeric material and an aqueous medium in contact with the housing. The aqueous medium includes i) a drug or an electrolyte salt or a mixture thereof and ii) a cetylpyridinium salt in an amount sufficient to inhibit microbial growth in the aqueous medium. The polymeric material is compatible with the cetylpyridinium salt. A process is also provided wherein when electric current flows from the source of electrical power so that the drug is transdermally delivered to the patient by electrotransport from the anodic reservoir, the cetylpyridinium salt is not transdermally delivered to the patient by electrotransport from the cathodic reservoir. A process for preparing a transdermal electrotransport drug delivery device is also provided.

Significant improvements in the utility of oxazolidone and polyoxazolidone compounds and in their manufacture can be achieved using a novel catalyst in the reaction between a hindered isocyanate and an epoxy, forming oxazolidone ring structures. The novel catalyst of the invention promotes the efficient and rapid formation of oxazolidone and polyoxazolidone compounds. The use of hindered isocyanate in the reaction prevents formation of side reaction products such as isocyanate trimer, allophanate and biuret.
