    摘要: An apparatus is described which is adaptable for use in coordinating UV/VIS absorbency values of a chromatography eluate containing antimicrobial constituents with bioautograph test results. Such an apparatus is comprised of a UV/VIS spectrophotometer equipped with a continuous recording monitor, a pair of parallel metering valves, a stream splitter, a rotatable drum adapted to hold a piece of absorbent material and means to rotate said drum. The method involves the use of such an apparatus in combination with the bioautograph test to coordinate said absorbency valves with the test results from a plurality of bioautograph test organisms.

    摘要: A motor driven inoculator is provided wherein a plurality of bacteriological transfer loops are mounted on a common head and are activated in response to the positioning of a tray of inoculum to cause the head to be lowered and then raised so that the loops pick up a sample of inoculum. The subsequent positioning of an antibiotic or antimicrobic microtube plate, etc. causes the head to be lowered and raised a second time so that the loops are lowered into the plate to place samples of inoculum in each of a plurality of wells in the plate by dispersion. The removal of the antibiotic plate initiates a sterilizing cycle wherein the head is lowered and raised a third time whereby the loops are heated to a very high temperature by passing electrical current therethrough to volatilize all material thereon and then they are subsequently air cooled prior to the beginning of the next cycle. In an alternative embodiment the inoculum on the loops may be dispersed by percussion.The inoculum tray includes a plurality of grooves and ridges so that a minimal amount of inoculum is necessary for use in placing samples on the loops by dipping.

    摘要: Bacteria of the genus Neisseria can be detected in a sample by separating a lysate of the sample into two parts, incubating one part with a solution of nicotinamide-adenine-dinucleotide (NAD) and 1,2-propanediol, incubating the other part with a solution of only the NAD, and then comparing the products of each incubation for reduction of NAD to NADH. A difference in NADH concentration indicates the presence of any enzyme specific to Neisseria, this enzyme having the capability of oxidizing 1,2-propanediol and reducing NAD. The structure of the enzyme is not completely understood but, because of those two characterizing properties, the nomenclature 1,2-propanediol dehydrogenase therefor is proposed herein.

    摘要: Cultures of microorganisms can be grown in a body cavity by situating therein a nutrient on a suitable carrier while maintaining the nutrient exposed to a predetermined gaseous atmosphere, so that microorganisms which require a particular atmosphere can grow directly in the body cavity on the nutrient. After a time sufficient for culture growth the carrier with the nutrient thereon are removed from the body cavity and inspected, for example, under a microscope with suitable dyes being used to stain the growth for rendering the latter easily visible. The carrier for the nutrient may take the form of a plastic sheet material having small protuberances which are thicker than a nutrient layer carried by the plastic sheet material and which serve to maintain apart from each other surfaces of the plastic and nutrient which otherwise might engage each other to prevent exposure of the nutrient to the predetermined atmosphere. The article is preferably packaged in a container which has in its interior sufficient moisture to prevent drying of the nutrient, the interior of the container being closed off from the outer atmosphere by the sheet material which is used for the container.
