摘要: Provided herein are synthetic peptides with enhanced antimicrobial and antibiofilm characteristics, and are biocompatible with mammalian cellular systems. The disclosed synthetic antimicrobial moieties include a mastoparan peptide having SEQ ID NO:1 and a pentapeptide motif formed from phenylalanine, leucine, proline, and two isoleucine residues, wherein the pentapeptide motif is conjugated the N-terminus of the mastoparan peptide. Also provided are compositions comprising the synthetic peptides, as well as methods of treating a microbial infection or removing a biofilm using the peptides.

摘要: The present invention relates to compositions and methods of use of anti-ADAMTS13 autoantibodies and fragments thereof. In one aspect, the invention includes a composition comprising an isolated anti-ADAMTS13 autoantibody or fragment thereof. In other aspects, methods are described for generating an in vivo model of thrombotic thrombocytopenic purpura (TTP) comprising introducing at least one anti-ADAMTS13 autoantibody or fragment thereof into a model organism and identifying an anti-autoimmune reagent for treating TTP.

摘要: Described is an assay termed Programmable Enzyme-Assisted Selective Exponential Amplification (PASEA) that concurrently amplifies both wild type and mutant alleles while selectively cleaving the former. With time, the rare mutant alleles dominate, and are readily detectable by direct detection, Sanger sequencing, and other readily available methods. Also described are point-of-care assays and microfluidic devices for performing PASEA.

摘要: A method of analyzing thoracic insufficiency syndrome (TIS) in a subject by performing quantitative dynamic magnetic resonance imaging (QdMRI) analysis. The QdMRI analysis includes performing four-dimensional (4D) image construction of a TIS subject's thoracic cavity. The 4D image includes a sequence of two dimensional (2D) images of the TIS subject's thoracic cavity over a respiratory cycle of the TIS subject. The QdMRI analysis also includes segmenting a region of interest (ROI) within the 4D image, determining TIS measurements within the ROI, comparing the TIS measurements to normal measurements determined from ROIs in 4D images of the thoracic cavities of normal subjects that are not afflicted by TIS, and outputting quantitative markers indicating deviation of the thoracic cavity of the TIS subject relative to the thoracic cavities of the normal subjects.

摘要: Compositions and methods relating to regulatable chimeric antigen receptors (RCARs), where the intracellular signaling or proliferation of the RCAR can be controlled to optimize the use of an RCAR-expressing cell to provide an immune response, are provided. For example, a RCAR can comprise a dimerization switch that, upon the presence of a dimerization molecule, can couple an intracellular signaling domain to an extracellular recognition element, e.g., an antigen binding domain, an inhibitory counter ligand binding domain, or costimulatory ECD domain. An RCAR can be engineered to include an appropriate antigen binding domain that is specific to a desired antigen target and used in the treatment of a disease.
