摘要: Genetically modified strains of Streptomyces tsukubaensis (S. tsukubaensis) can be used for an improved fermentation process for the preparation of tacrolimus or a salt or derivative thereof by cultivation of these genetically modified strains. Novel genes allowing biosynthesis of allylmalonyl-CoA can be used for polyketide production with allylmalonyl extender unit.

One important area is the isolation and potential use of novel FKBP-ligand analogues and host cells that produce these compounds. The invention is particularly concerned with methods for the efficient transformation of strains that produce FKBP analogues and recombinant cells in which cloned genes or gene cassettes are expressed to generate novel compounds such as polyketide (especially rapamycin) FKBP-ligand analogues, and to processes for their preparation, and to means employed therein (e.g. nucleic acids, vectors, gene cassettes and genetically modified strains).

摘要: A bioactive compound, or family of compounds, is biosynthesised by a cell using a plurality of enzymic activities. Host cells are provided that substantially lack the bioactivity, but possess at least a first one of the enzymic activities (or nucleic acid encoding a corresponding enzyme). The cells are transformed with nucleic acid expressible to provide at least a second enzymic activity which enables the cells to produce a bioactive compound. A family of cells may be transformed with a multiplicity of different nucleic acids, leading to a library of cells including cells producing different bioactive product, and cells not producing bioactive products. Transformed cells may be screened for bioactivity. Active cells may be isolated and cultured, and bioactive compounds may be isolated.
