This invention relates to a method of producing a taxane-type diterpene(s) wherein tissues or cells of a plant which produces taxane-type diterpene(s) is cultured in the presence of at least one selected from the group consisting of jasmonic acids, compounds containing a heavy metal, complex ions containing a heavy metal, heavy metal ions, amines and antiethylene agents, a method of producing a taxane-type diterpene wherein the tissues or the cells of the plant are cultured by controlling the oxygen concentration in a gas phase in a culture vessel to less than the oxygen concentration in the atmosphere from the initial stage of the culture, or by controlling the dissolved oxygen concentration in a fluid medium which is in contact with the tissue or the cell to less than the saturated dissolved oxygen concentration at that temperature from the initial stage of the culture, a method of producing a taxane-type diterpene wherein the tissue or the cell of the plant is cultured in a culture vessel, while oxygenic gas containing 0.03-10% of carbon dioxide is used as aeration gas to be introduced to the vessel, and a method of obtaining highly productive cultured cells for the taxane-type diterpene wherein cultured cells of the plant which produces the taxane-type diterpene are separated into a plurality of layers according to the difference in their specific gravities, and the cells contained in at least one layer are cultured, then highly productive cultured cells for the taxane-type diterpene are selected from among those cultured cells.The present invention allows industrial production of a taxane-type diterpene such as taxol which is useful as a therapeutic agent for cancer.

Methods for isolating polynucleotides encoding antibodies against lesional tissues are provided, wherein the methods comprise the steps of: (a) isolating a B cell(s) that infiltrates into a lesional tissue of interest; and (b) obtaining an antibody-encoding polynucleotide from the isolated B cell(s). The lesions may be a cancer tissue or such. Antibody genes can be obtained without depending on B cell cloning. Accordingly, it is also possible to obtain genes encoding human-derived antibodies which are difficult to clone. Genes that encode antibodies against cancer can be obtained using cancer tissues as the lesion.

The present invention discloses a method for producing a taxane-type diterpene(s) by culturing cells of a taxane-type diterpene-producing plant, comprising one or both of the the following steps (a) and (b) to increase the ratio of those cell clusters suitable in size for diterpene production: (a) at least one operation for removing large cell clusters is conducted with a sieve and/or a filter during preculture of the cells to be supplied to main culture for the production of taxane-type diterpenes or at the time of cell transfer from one preculture to the subsequent preculture; (b) the cells are cultured under strong agitation conditions.

It is intended to find characteristics of a pectin having a high molecular weight as compared with the existing pectins which are obtained by extracting plants and provide a method whereby the pectin can be supplied in a large amount. A callus derived from a plant is cultured so as to produce a pectin having a high molecular weight in the culture or the culture broth. Then the desired pectin is obtained by separating from the thus obtained culture or culture broth and purifying. The pectin thus obtained of the present invention has favorable properties as a moisturizer and, therefore, is excellent as a cosmetic material.
