Abstract:
The present invention relates to a novel mutant of L-asparaginase enzyme characterized in having high thermostability, pH stability and no glutaminase activity useful for therapeutics and the process of preparing the same. The present invention specifically relates to mutant's MTCC 5580, MTCC 5581 and MTCC 5582 characterized in having higher stability, no glutaminase activity etc., to allow their usage in the form of improved protein therapeutics. A thermostable L-asparaginase from P. furiosus was cloned and expressed in E. coli host. The enzyme was engineered at its active site to create three different mutants based on structural and sequence analysis with a E. coli—derived enzyme homologue. The mutants MTCC 5580, MTCC 5581 and MTCC 5582 were tested for their stability, substrate affinity, optimum pH and temperature of activity and cytotoxicity. Based on the studies, all the three enzymes were found thermostable and with no glutaminase activity as compared to other available enzyme EcA II. MTCC 5579 and the above said three mutants showed the cytotoxicity on the leukemic cell lines. The present study showed that these enzymes are promising candidates for the treatment of leukemia.

Systems and methods for determining and providing call disposition information for outgoing calls are provided. More particularly, the disposition of a call placed from a communication device is determined, and associated with a call log for that communication device. The call disposition information is displayed or otherwise output to a user when the call log is accessed by the user. The call disposition information can be determined by a call disposition application running on a communication server that provides the call disposition information to a communication device for display by that communication device.

A system and method are disclosed relating to authenticating software licenses associated with a software product. During an installation process, a purchaser of a software product may contact a vendor representative via a telephone connection and verbally relay a data packet, which may be an installation ID. The vendor server creates a reversible hash from the data packet, a known seed value and additional information relating to the software product or license. The server encrypts the hash to generate a confirmation ID, and sends the confirmation ID to the software product purchaser.

The invention features a method of producing a high mannose glucocerebrosidase (hmGCB) which includes: providing a cell which is capable of expressing glucocerebrosidase (GCB), and allowing production of GCB having a precursor oligosaccharide under conditions which prevent the removal of at least one mannose residue distal to the pentasaccharide core of the precursor oligosaccharide of GCB, to thereby produce an hmGCB preparation. Preferably, the condition which prevents the removal of at least one mannose residue distal to the pentasaccharide core is inhibition of a class 1 processing mannosidase and/or a class 2 processing mannosidase. The invention also features an hmGCB preparation and methods of using an hmGCB preparation.
