Compositions and methods for dual imaging and for dual chemotherapy and radiotherapy are disclosed. More particularly, the invention concerns compounds comprising the structure X1-Y-X2, wherein Y comprises two or more carbohydrate residues covalently attached to one another, X1 and X2 are diagnostic or therapeutic moieties covalently attached to Y, provided that when Y does not comprise a glucosamine residue, X1 and X2 are diagnostic moieties. The present invention also concerns methods of synthesis of these compounds, application of such compounds for dual imaging and treatment of hyperproliferative disease, and kits for preparing a radiolabeled therapeutic or diagnostic compound.

The present invention provides automated systems for the purification of radioisotopes, the formulation of radiopharmaceuticals or both. In preferred embodiments the radioisotopes are produced by a generator. In further preferred embodiments the automated systems comprise replaceable cassettes for easy replacement of the system's networks of tubing. Also provided are replaceable cassettes and kits for use with the automated systems of the invention.The invention further provides methods of using an automated system to purify radioisotopes, formulate radiopharmaceuticals or to do both in series.

Novel compositions for imaging that include (a) a polypeptide that includes two or more consecutive amino acids that will function to non-covalently bind valent metal ions and (2) a valent metal ion chelated to at least one of the two consecutive amino acids, are disclosed. Also disclosed are methods of imaging using these novel compositions, such as methods of imaging a tumor within a subject. Methods of synthesizing an imaging agent and kits for preparing an imaging agent are also disclosed. Methods for determining the effectiveness of a candidate substance as an imaging agent that involve conjugating or chelating the candidate substance with a polypeptide that includes two or more consecutive amino acids that will function to non-covalently bind valent metal ions.

A hydrogel composition is generated in situ in the tumor by administering a polymer, such as a polysaccharide or a polyamino acid, with a therapeutic agent, such as a radionuclide or a drug, and administering a cross-linking agent. The hydrogel/therapeutic agent composition is retained in the tumor for safe and efficient tumor therapy. Alternatively, a hydrogel composition is generated in situ in an artery which nourishes a tumor to occlude the artery.

Systems and methods are described for efficiently producing a radiopharmaceutical agent. The system includes a generator that provides an admixture comprising hydrochloric acid and gallium-68 (Ga-68). A heater initiates a removal process by evaporating the hydrochloric acid from the admixture resulting in a substantially purified Ga-68. A plurality of valves provides a buffer and a prodrug that gets mixed with the Ga-68 to produce Ga-68 radiopharmaceutical agent. Additionally, the valves may provide water or a transchelator to the Ga-68 radiopharmaceutical agent to optimize the yield. A method includes providing an admixture comprising hydrochloric acid and Ga-68. The method also includes removing the hydrochloric acid by heating the admixture and subsequently evaporating the hydrochloric acid, resulting in a substantially purified Ga-68. The method further includes adding a buffer and a prodrug to the substantially purified Ga-68 to produce the Ga-68 radiopharmaceutical agent.
