摘要: This invention relates to isolated polynucleotides encoding WUS polypeptides. The invention further provides isolated WUS polypeptides. The invention also provides methods of using the polynucleotides to modulate the level of WUS, improve transformation efficiency, to stimulate plant cell growth, including stem cells, to stimulate organogenesis, to stimulate somatic embryogenesis, to induce apomixis, and to provide a positive selection for cells comprising the polynucleotide. The invention also relates to cells, plants and seeds comprising the polynucleotides of the invention or produced by the methods of the invention.

The invention also relates to the construction of a chimeric gene encoding all or a portion of the WUS protein, in sense or antisense orientation, wherein expression of the chimeric gene results in production of altered levels of the WUS protein in a transformed host cell.

摘要: Compositions and methods are provided for the targeted integration of a polynucleotide sequence of interest into the genome of a plant or plant cell. The methods and compositions employ recognition sites for endonucleases and endonucleases in combination with site-specific recombination sites/recombinases to provide an effective system for establishing target sites within the genome of a plant, plant cell or seed. Once such target sites are established, a variety of methods can be employed to further modify the target sites such that they contain a variety of polynucleotides of interest.

摘要: Compositions and methods for the efficient co-transformation of a plant are provided. Novel compositions are Agrobacterium strains that have been engineered to comprise at least two binary vector plasmids in addition to a helper plasmid comprising the vir functions. Each of the binary vectors comprises its own T-DNA borders flanking a heterologous nucleotide sequence of interest. Methods of the invention comprise the use of these novel multiple-binary vector Agrobacterium strains to co-transform a plant. In this manner, heterologous nucleotide sequences of interest residing on different binary vectors can be independently introduced into the plant in a single transformation event and incorporated in the plant's nuclear DNA in an unlinked manner. The invention also provides for regenerated, fertile transgenic plants, transgenic seeds produced therefrom, and T1 and subsequent generations.
