Abstract: The disclosure provides variant Staphylococcus aureus Cas9 (SaCas9) proteins with altered specificity for protospacer adjacent motif (PAM) sequences. The disclosure also is directed to CRISPR/Cas9 systems and methods of altering a genomic DNA sequence using the variant SaCas9 protein. Methods of generating variant Cas9 proteins with altered PAM specificity are also disclosed.
