Abstract: Compositions, methods, and kits are provided for diagnosing and treating uveitis. In particular, biomarkers have been identified that can be used to distinguish infectious uveitis from noninfectious uveitis, and further discriminate among bacterial, viral, and fungal uveitis. These biomarkers can be used alone or in combination with one or more additional biomarkers or relevant clinical parameters in prognosis, diagnosis, or monitoring treatment of uveitis.

Abstract: Methods are provided for treating and diagnosing diseases and disorders associated with retinal degeneration, such as retinitis pigmentosa. Dietary supplementation with specific metabolites and vitamins can prolong vision and provide a neuroprotective effect. In particular, dietary supplementation with α-ketoglutarate, or a derivative thereof, significantly prolongs photoreceptor cell survival and visual function. In addition, dietary supplementation with B vitamins and a ketogenic diet also improves photoreceptor cell survival and delays disease progression in some cases. Additionally, compositions, methods, and kits are provided for diagnosing a subject with retinitis pigmentosa based on expression levels of vitreous biomarkers.

Abstract: Compositions, methods, and kits are provided for assaying calpain-5 activity and inhibition. In particular, novel peptide substrates are provided for detecting calpain-5, measuring calpain-5 activity, and screening for inhibitors of calpain-5 to identify potential therapeutic agents for treating retinal diseases and other diseases associated with calpain-5 hyperactivity. Additionally, novel inhibitors of calpain-5 are also provided.

Abstract: A high-resolution crystallographic structure of the mutant human G267S calpain-5 protease core domain at 2.22 Å resolution is provided. The G267S mutation is associated with hyperactivity of calpain-5 and is linked to the inherited disease, neovascular inflammatory vitreoretinopathy. Methods of using the crystallographic structure in rational design of small molecule drugs that inhibit calpain-5 for treatment of retinal diseases and other diseases associated with calpain-5 hyperactivity are also provided.

Abstract: A construct comprising a promoter specific for non-pigmented ciliary epithelial cells (NPCECs) is provided. In particular, the construct comprises a BEST2 minimal promoter that confers NPCEC-specific expression. The BEST2 minimal promoter may be operably linked to an expressible sequence such as a gene encoding a polypeptide of interest, a regulatory RNA sequence, a reporter gene, and the like. Such constructs may be provided in an expression vector, for example, with the BEST2 minimal promoter operably linked to an expressible sequence or in a host cell genetically modified with such an expression vector.
