An apparatus for detecting an object capable of emitting light. The apparatus includes a light source (102) and a waveguide (110). The waveguide (110) includes a core layer (112) and a first cladding layer (114). At least one nanowell (120) is formed in at least the first cladding layer (114). The apparatus further includes a light detector (106). The light detector (106) can detect light emitted from a single molecule object contained in the at least one nanowell (120).

A single-molecule detection system and a plurality of methods of detecting a single-molecule object or sequencing a nucleic acid are provided. The single-molecule detection system comprises: a movable light coupler (100), a waveguide (110) and a light detector (102). The waveguide (110) comprises: a core layer (112) and a first cladding layer (114), wherein at least one adapter site (104) for the movable light coupler (100) is formed in at least the first cladding layer (114). In the methods, the detection system is used to detect a single-molecule object or sequence a nucleic acid.

The apparatus comprises a light detector comprising at least two optical sensors capable of determining the intensity of the light; and a computer processing output signal generated by the optical sensors and comparing a processing result with a known result corresponding to a know type to determine whether the object belongs to the known type. A method for detecting an object capable of emitting light is also provided.
