Preparation of biologically active tropane derivatives of formula (1) and structural isomers thereof wherein R equals C1 to C8 alkyl and Ar is an aromatic ring moiety. The compounds are prepared by decomposing vinyl-diazomethanes in the presence of pyrroles to provide a bicyclic ring containing the basic tropane ring system which is then reacted with an aryl Grignard reagent in the presence of a copper (I) or (II) salt to provide 3-aryl-tropane derivatives.

Detectably-labeled compounds of formula (I) wherein A, B, C, D, E, F, R, X, Y and Z have any of the meanings given in the specification, and their pharmaceutically acceptable salts, are useful in competitive assays to assay for the presence of sigma 2 receptors. Also disclosed are pharmaceutical compositions containing such a compound of formula (I), methods for their use and intermediates useful for preparing such compounds of formula (I).
