The present invention is directed to a process for making homogeneous, thermoreversible gel films comprising the steps of: (i) heating, hydrating, mixing, solubilizing, and, optionally, de-aerating a high solids, low moisture film forming composition in an apparatus providing sufficient shear, temperature and residence time to form a homogeneous molten composition, wherein the temperature is at or above the solubilizing temperature of said composition; (ii) feeding the molten composition into at least one of a mixer, pump or devolatilizer; and (iii) cooling the homogeneous molten composition at or below its gelling temperature to form said gel films. The present invention is also directed to various products made from such films, such as the gel films themselves, soft capsules, solid dosage forms and delivery systems.

A method for the preparation of microcrystalline cellulose containing tablets by roller compaction followed by tabletting is disclosed. A tablet formulation is converted to a dry granulate by roller compaction, and the dry granulate lubricated dry granulate and compacted to a tablet. The tablet formulation comprises at least one active, an microcrystalline cellulose containing material, and, optionally other pharmaceutically acceptable excipients. The microcrystalline cellulose containing material has a maximum primary compaction tensile strength of at least 9 MPa or at least 9.5 MPa and a secondary compaction tensile strength of at least 5 MPa, at least 5.5 MPa, or at least 6 MPa.

Abstract:
The invention relates to composites comprising a polysaccharide gelled within pores of a foam, methods of preparation, and uses thereof, for example, in biomedical applications such as cell culture media and implants, controlled release delivery systems, food applications, industrial applications, and personal care applications such as cosmetic and oral hygiene. The composites of the present invention are simple to formulate using few steps and are useful for entrapping heat-sensitive components, such as cells, drugs, flavors or fragrances within the polysaccharide gel. In addition, the invention provides for a composite able to gently immobilize fragile components, such as living cells, without exposing such components to shear forces.

A fluid mixing apparatus for mixing fluids with steam to form gels, gel films, films and/or shaped articles includes a steam inlet for injecting steam into a mixing zone, and multiple fluid inlets for separately delivering incompatible fluids to the mixing zone. As an example, a first fluid may be a solution of one or more starches and water, and the second fluid may be a solution of one or more plasticizers and one or more hydrocolloids. Upon mixing with steam, the resulting flowable molten mass or slurry mixture can be formed into a homogeneous film that is suitable for producing soft capsules, hard capsules and other film products, or formed into a shaped article.
