The sample preparation device, such as a standard pipette tip, has chromatography media bound to the inside surface of the tip so as to create a monolayer of particles that are readily accessible to fluids that flow through the pipette tip. The device can be used for washing, protein binding, elution processes, etc. A method of sample preparation using the device of the present invention is also disclosed.

The method begins with starting components or a ligand library and provides methods of creating catalyst or product libraries, which are then tested in a reaction of interest. The system uses methods of robotic handling for moving libraries from station to station. The method and apparatus are especially useful for synthesizing, screening, and characterizing combinatorial catalyst libraries.

The chamber also includes a centering collar (31) with an opening through which the capillary passes to center the capillary in an outlet opening (33) of the chamber element (22). The capillary may also include a bead (39) to control the extension through the opening (33). The system also includes a heater (32) extending along the length of the chamber which heats the sheath gas, which in turn heats the capillary along most of its length.

Heater wire (160) is similarly wrapped with a ceramic fiber electrical insulating layer (158). Resistive temperature device (RTD) wire (154) is positioned contiguous an outer surface of insulating layer (156). The composite assembly including insulated capillary GC column (152), insulated heater wire (160), and RTD wire (154), are bound together by applying thereto a spiral wrapped ceramic fiber insulating layer (162).

Disclosed is a method for increasing the electroosmotic flow rate available for a silica surface. In the method, there is provided an electrophoretic channel which is defined by one or more silica surfaces. The surface(s) are contacted with an alkaline aqueous solution containing a solubilized silicate-monovalent metal complex in an amount effective to increase the acidity of the silica surfaces(s), as evidenced by a reduction in the average bulk pKa of the surface(s). The achieved increase in acidity is greater than would be obtained using an otherwise identical solution lacking said silicate. In one preferred embodiment, the monovalent metal used in the solution is Li?+, Na+, or K+¿. Also disclosed is a method for increasing the acidity of a silica surface, by contacting the surface with an alkaline aqueous solution of the type noted above.

The liquid chromatograph comprises a separation column (10), a buffer pump (7) for feeding a plurality of buffers 1, 2, 3 and 4 into the separation column (10), a sampler (9) disposed in a flow path between the buffer pump (7) and the separation column (10) for introducing a sample to be analyzed into the flow path, and a reaction coil (14) for mixing and then reacting amino acids separated by the separation column (10) with a ninhydrin reagent. The separation column (10) is sized so that a ratio (L1/R) of its length L1 to its inner diameter R is not more than 10. The particle size of ion exchange resin filled in the separation column (10) is selected to be not more than 4 µm.

Devices formed according to the invention include associated laser-ablated features required for function, such as analyte detection device (34, 36) and fluid communication device (10). Miniaturized columns constructed under the invention find use in any analysis system performed on either small and/or macromolecular solutes in the liquid phase and may employ chromatographic, electrophoretic, electromatographic separation device, or any combination thereof.
