    摘要: A method and a device for separating a component, such as fibrin monomer from blood, by centrifugation, involve feeding of blood admixed an anticoagulant to a first annular chamber in a device, where the annular chamber is defined by a cylindrical outer wall and a cylindrical inner wall, both walls extending coaxially about a common axis, as well as by a top wall and a bottom wall. The top wall or the bottom wall is formed by a piston body displaceable within the first chamber. This method involves furthermore a centrifugation of the device about the said common axis to substantially separate blood into a cell fraction and a plasma fraction followed by the resulting plasma fraction being transferred while influenced by the piston body to a second chamber defined by an outer cylindrical wall. The outer cylindrical wall extends coaxially with the said common axis, whereby a fraction with fibrin I is caused to be separated in the second chamber while a suitable enzyme is being added. The separation of fibrin monomer from the plasma fraction in the second chamber is carried out during continued centrifugation whereby a fibrin polymer is deposited on the cylindrical outer wall of said second chamber, whereafter the fluid fraction collected at the bottom of the second chamber is transferred while influenced by the piston body to the first chamber. The fraction with fibrin polymer remaining in the second chamber is substantially deposited on the cylindrical wall and is caused to be dissolved by addition of a solvent and by centrifugation, whereafter it is transferred to a receiving container by passing after addition of a binder to the enzyme through a filter removing said enzyme, whereby a fibrin monomer containing solution is provided.

    摘要: A method for separating a blood component, e.g., fibrin monomer, from blood or plasma. The method utilizes a container with a reaction chamber for receiving the plasma. The reaction chamber is defined by a wall and an agent is supplied to the reaction chamber for converting the fibrinogen content of the plasma into a non-cross-linked fibrin polymer. The method also utilizes a device for centrifuging the reaction chamber with the plasma and the agent to a degree sufficient for separating the non-cross-linked fibrin polymer from the plasma, for depositing the polymer on the outer wall of the reaction chamber, and for expelling the remaining plasma from the reaction chamber. A solvent is supplied to the reaction chamber for dissolving the non-cross-linked fibrin polymer. The solvent is supplied to the reaction chamber in a first amount for dissolving the non-cross-linked fibrin polymer which is less than the total amount of solvent for dissolving the polymer. The remaining solvent is supplied to the reaction chamber in one or more portions to flush the reaction chamber and other portions of the device and recover additional non-cross-linked fibrin polymer.

    摘要: An instrumentality for promulgating the cryoprecipitation of fibrinogen from a blood product. The instrumentality contemplates a container for the blood product, an apparatus for creating the fibrinogen within the container and a method of manipulating the container within the apparatus and subsequently after fibrinogen has been formed. The apparatus includes a receiver within which the container is supported, a motion transfer device for the receiver to impart motion to the container while simultaneously subjecting the container to a temperature differential to cause heat transfer. The motion imparted to the container results in a thin coating of the blood product being disposed on an interior surface of the container which, in turn, is exposed to a heat transfer fluid through the wall of the container. Successive coatings placed on an interior of the container are timed such that each coating is placed on a previous coating that has changed phase. Flexure of the container after the contents of the container have been frozen can occur by providing a vacuum within the container causing an implosion of frozen material. Subsequent phase change back to a liquid and then a second freeze/thaw followed by centrifugation causes a cryo-fraction to be precipitated therefrom. The container within which the blood product is housed includes an opening for admission of the blood product therewithin, an outlet for extraction of fibrinogen and access to a source which can provide a pressure differential vis a vis the interior and exterior of the container. Information of the temperature profile within the container may also be provided.
