摘要: A method and tube for separating a sample. The tube is configured to separate agglomerated or clumped material or other sample components having over a particular size from other smaller size portions of the sample. For example, blood clots may be separated from serum of a blood sample. The tube includes a chamber with a closed bottom and a sidewall extending upwardly from the bottom, and a sample inlet branch coupled to the chamber. A slit is positioned between the chamber and the sample inlet branch so that after a sample is placed into the sample inlet branch, a portion of the sample having a size smaller than the slit (e.g., liquid material) passes through the slit and into the chamber, and material in the sample having a size larger than the slit remains in the sample inlet branch.

摘要: Blood separation systems and methods are provided for separating blood under conditions of reduced plasma clarity. The system may assess plasma clarity by monitoring light transmissivity of plasma or comparing an actual plasma flow rate to an ideal plasma flow rate, with a low flow rate indicating decreased clarity, which may be addressed by increasing the plasma flow rate. For extracorporeal photopheresis, plasma clarity may be a factor in determining the dosage of irradiating light to apply to mononuclear cells. A fluid processing assembly for mononuclear cell collection may include visual indicium, which an operator may use to determine when to end mononuclear cell collection. The system may detect red blood cells flowing toward a mononuclear cell collection container and reverse the direction of flow to prevent red blood cells from entering the container. An operator may also be enabled to selectively begin and/or end harvesting of mononuclear cells.

Thereby, a blood collection unit rapidly providing coagulated blood products is provided. Said blood products are separated effectively into blood serum and a blood clot absent of additives and contaminants which would make the blood unsuited for certain uses, such as blood testing/analysis, and LeucoPatch®.

摘要: The present specification discloses an automated method and apparatus useful for separating and harvesting cells of interest, e.g., mononuclear cells, in a whole blood sample. The method of the invention uses the aspirating/dispensing probe of an automated sample preparation instrument to underlay a density gradient medium beneath a whole blood sample in a centrifugation tube, and the same probe is used to harvest cells of interest from a cell layer formed in the tube as a result of a centrifugation step. In harvesting cells, the probe is advanced inside the tube by a fixed, predetermined distance at which the probe tip (i.e., its aspiration port) is known to be located at, or within a predetermined distance below, the bottom of the cell layer. A predetermined volume of liquid is then aspirated through the probe tip, whereby most cells of interest (and more than 90% of those cells that can be harvested by a flawless manual method) are removed from the cell layer and collected for analysis. Preferably, the probe is caused to move laterally with respect to the tube during the aspiration of the cell layer, whereby cells offset from the center of the container are readily harvested.
