The apparatus comprises a sample collection and analysis system used to collect a sample, mix reagents, react the sample, and collect it in a measurement chamber. The system includes an instrument having a photon detection assembly for use with the sample testing device and one or more probe assemblies that optically cooperate with the instrument. The instrument includes a dark chamber with a reflective interior surface which may be concave or preferably spherical, and a photon detection sensor such as a multi-pixel photon counter sensor. A substantially transparent portion of the probe assembly, and liquid contained therein, focus bioluminescence toward the photon detection sensor.

An integrated biosensing device detects emissions from a sample when illuminated. A photo detector (20) is adjacent a site for retaining the sample (40) or receiving excitation radiation for impingement on the sample (40). A reflector (10) deflects the illumination onto the sample site and substantially guides the excitation radiation away from the photo detector. By providing a reflector, a ratio of desired detection of emissions to unwanted detection of the illumination light can be improved. This can be achieved by a reduction in an amount of the illumination reaching the photo detector, and/or by an increase in the amount of illumination of the sample and thus in the amount of emissions reaching the detector. This can be achieved more cost effectively than by using a filter. The illumination can be from above or below if the substrate is transparent.

A parallel-beam optical tomography system for imaging an object of interest includes a parallel ray beam radiation source that illuminates the object of interest with a plurality of parallel radiation beams. After passing through the object of interest the pattern of transmitted or emitted radiation intensities is magnified by a post specimen optical element or elements. An object containing tube is located within an outer tube, wherein the object of interest is held within or flows through the object containing tube. A motor may be coupled to rotate and/or translate the object containing tube to present differing views of the object of interest. One or more detector arrays are located to receive the emerging radiation from the post specimen magnifying optic. Two- or three-dimensional images may be reconstructed from the magnified parallel projection data.
