The present invention relates to the construction and engineering of cells, more particularly microorganisms for producing PUFAs with four or more double bonds from non-fatty acid substrates through heterologous expression of an oxygen requiring pathway. The invention especially involves improvement of the PUFA content in the host organism through fermentation optimization, e.g. decreasing the temperature and/or designing an optimal medium, or through improving the flux towards fatty acids by metabolic engineering, e.g. through over-expression of fatty acid synthases, over-expression of other enzymes involved in biosynthesis of the precursor for PUFAs, or codon optimization of the heterologous genes, or expression of heterologous enzymes involved in the biosynthesis of the precursor for PUFAs.

Disclosed is a method for producing a DHA phospholipid comprising an ω3 unsaturated fatty acid, particularly DHA, as a constituent lipid by using a microorganism in a simpler manner. Specifically disclosed is a method for producing a phospholipid comprising an ω3 unsaturated fatty acid as a constituent lipid, which comprises the steps of: growing a microorganism capable of producing the ω3 unsaturated fatty acid in a culture medium containing a carbon source; and further culturing the grown microorganism in a culture medium without any carbon source. The method enables to produce a highly value-added phospholipid which comprises an ω3 unsaturated fatty acid as a constituent lipid by using a microorganism capable of producing the ω3 unsaturated fatty acid in a large quantity.

A process for the production of phospholipids by transphosphatidylation, which comprises the step of mixing a starting phospholipid with a receptor having one or more hydroxyl groups, phospholipase D and water in the absence of an organic solvent and homogenizing the resulting mixture and the step of subjecting the obtained homogeneous mixture to a reaction at 15 to 65 DEG C. The homogeneous mixture has a lamellar lyotropic liquid crystal structure and can give an objective phospholipid via transphosphatidylation without using an organic solvent or calcium.
