At least one of the plurality of struts includes at least one opening (24, 26) extending at least partially through a thickness of said strut. A beneficial agent is loaded into the opening within the strut in layers to achieve desired temporal release kinetics of the agent. The beneficial agent can be a chemically ablative agent use to create a lesion to treat atrial fibrillation. A wide variety of delivery profiles can be achieved including zero order, pulsatile, increasing, decrease, sinusoidal, and other delivery profiles.

A medical device for implantation into vessels or luminal structures within the body is provided. The medical device, such as a stent and a synthetic graft, is coated with a pharmaceutical composition consisting of a controlled-release matrix and one or more pharmaceutical substances for direct delivery of drugs to surrounding tissues. The coating on the medical device further comprises a ligand such as an antibody or a small molecule for capturing progenitor endothelial cells in the blood contacting surface of the device for restoring an endothelium at the site of injury. In particular, the drug-coated stents are for use, for example, in balloon angioplasty procedures for preventing or inhibiting restenosis.

The present invention provides inflatable porous implants, such as grafts, stent-grafts, and bladders, as well as methods and kits for drug delivery. In particular, the grafts and stent-grafts of the present invention provide for the delivery of a therapeutic agent into a flow conduit in the body. The inflatable porous implants provide for direct delivery of larger, more precise dosages of drugs over longer administration periods into the body. Moreover, these inflatable porous implants are often flexible when inserted and have a low profile delivery configuration for easy placement. The implants of the present invention further provide a mechanical or structural function in addition to drug delivery in a single integrated structure.

The apparati include microchip devices whcih have reservoirs containing the molecules for release. The apparati and methods provide for active orpassive controlled relase of the molecules. Preferred embodiments include systems for intravenous administration of drugs, wherein drug molecules are released from the microchip devices into a carrier fluid ex vivo, such as a saline solution, forming a drug/saline solution mixture which is then delivered to a patient intravenously.

An implantable medical device (10) such as a catheter with a pharmacologically active ingredient layered between outer and inner elongated member tubes (11 and 13) for minimizing the risk of infection or other physiological afflictions associated with the implantation thereof. The outer and inner elongated member tubes include a base material mixed with a bioactive material including, for example, one and/or the other of rifampin and minocycline. An itermediate tube or layer (18) is positioned between and in communication with the outer and inner elongated member tubes (11 and 13). The intermediate tube or layer also includes a base material of, for example, silicone with a pharmacologically active ingredient mixed therein. The slower diffusion rate minocycline is included as or part of the bioactive material in the base material of the outer and inner elongated member tubes. The pharmacologically active ingredient or bioactive material such as a mixture of rifampin and minocycline is included in the base material of the intermediate tube or layer, which permeates through the inner and outer member tubes and diffuses therefrom or therethrough for treating tissues surrounding the catheter concomitantly.
