Disclosed is a glycosaminoglycan-polycation complex as a matrix material for use in tissue regeneration, such as cartilage repair, and a preparation method thereof. The complex is synthesized by crosslinking glycosaminoglycan and polycation with a polyfunctional crosslinking agent under physiological conditions. In this method, the crosslinking agent consists of polyethyleneglycol having two or more electrophilic leaving groups (e.g. succinimidyl group or its derivatives) at the carboxyl terminal thereof, and the concentration of the crosslinking agent is in the range of 0.3 to 3 mM. According the present invention, the crosslinking reaction can be performed under physiological conditions to avoid conventional problems of adverse affect on cells (cell death) and the formation of a polyion complex (inhomogeneous precipitate) due to a crosslinking reaction in alcohol or water. Thus, the crosslinking reaction can be conducted in the presence of cells mixed to the glycosaminoglycan and polycation in advance.

A medical and dental curable material is disclosed, which contains at least one of α-tricalcium phosphate and tetracalcium phosphate as a necessary ingredient. Conventional medical and dental curing materials containing α-tricalcium phosphate and/or tetracalcium phosphate as a necessary ingredient are unsatisfactory as to kneading workability and curing time since an aqueous solution of a single kind of organic acid is used as a curing agent. The medical and dental material of the present invention, which is satisfactory as to both kneading workability and curing time, uses as a curing-adjusting agent (1) tannin, (2) tannin and collagen, (3) collagen and organic acid, (4) tannin, collagen and organic acid, (5) tannin and organic acid or (6) at least two organic acids.

Therefore, it is feared that an injectable crosslinked biological substance might exert an undesirable effect (cell death) on cells and tissues. Moreover, a polyion complex (a non-homogeneous precipitate) is formed in this case. A glycosaminoglycan-polycation complex is formed by crosslinking glycosaminoglycan and polycation under physiological conditions with the use of 0.3 to 3 mM of a crosslinking agent which has two or more electrophilic leaving groups (for example, succinimidyl group or its derivative) at the carboxyl end of polyethylene glycol. Since the synthesis can be carried out under physiological conditions, the crosslinking can be achieved by preliminarily mixing the cells.

The object of the present invention is to provide: a hybrid material for regeneration of living body tissue, which can retain sufficient mechanical strength for a term needed to regenerate the tissue, and which simultaneously has moderate biodegradation absorbability which enables degradation and disappearance rapidly after the regeneration of the tissue. As means for solving the problems, the hybrid material for regeneration of living body tissue, according to the present invention, comprises a naturally-occurring polymer and a biodegradation-absorbable synthetic polymer, wherein the biodegradation-absorbable synthetic polymer exists in a fibrous form in a sponge including the naturally-occurring polymer, and wherein the naturally-occurring polymer is crosslinked.
