There is disclosed a recombinant DNA plasmid comprising a vector and a promoter of a nitrogenase complex gene from a Rhizobium japonicum bacterium. The plasmid may additionally comprise a structural gene (which may be a gene from a Rhizobium species or a foreign gene) inserted in such orientation and location as to be expressible under control of the promoter. Also disclosed are the nucleotide sequences of nifH and nifD promoters which may be used in the plasmid of the invention.

A novel method of producing a plant with a marker closely linked to a target locus, in particular a nuclear male sterile target locus, is described. The method involves transformation of a group of plants in order to introduce a marker into each plant, and isolation of a plant with the marker closely linked to a target locus. The markers include visible markers and dominant conditional lethal markers. The method is of particular use for hybrid seed production where the target locus is a nuclear male sterile locus.

The invention relates to a process for the incorporation of foreign DNA into the genome of dicotyledonous plants by infecting the plants or incubating plant protoplasts with Agrobacterium tumefaciens bacteria, which contain one or more Ti (tumour inducing) plasmids, wherein as Ti plasmid a stable cointegrate plasmid composed of the plasmid R772 and the plasmid pTiB6 with foreign DNA incorporated in the T-region of the Ti component of the cointegrate plasmid is applied as well as to a cointegrate plasmid pAL969 and cointegrate plasmids derived from the cointegrate plasmid by the incorporation of foreign DNA into the T-region of the Ti component.

Plant protoplasts are exposed for interaction with an external agent by exposing the plasma membrane of protoplasts in the root hairs of the plant without release of protoplasts from the plant. This may be achieved by enzymatically degrading the plant cell wall atthe apices of root hairs under conditions which expose the plasma membrane whilst maintaining or readily permitting restoration of the essential functional integrity of the plant. Enzymatic degradation of the cell wall of the apices of root hairs is possible in a wide range of crop species and other species and to expose the plasma membrane with partial protoplast release. This enables direct interaction with plasmids, viruses and micro-organisms at the surface of the plasma membrane of their exposed protoplasts, or the fusion of these with isolated protoplasts of other plant systems.

Enhanced resistance to glyphosate, an inhibitor of the aromatic amino acid biosynthesis pathway, is imparted to a glyphosate sensitive host A mutated aroA gene is employed which expresses 5-enolpyruvyl-3-phosphoshikimate synthetase (EC: 2.5.1.19) (ES-3-P synthetase). Methods are provided for obtaining the aroA mutation which provides the enzyme resistant to inhibition by glyphosate, means for introducing the structural gene into a sensitive host, as well as providing a method of producing the enzyme. The method of construction of at least a portion of, an RNA vector from an RNA plant virus, e.g. tobacco mosaic virus (TMV) using as specific functional regions, at least two nucleotide sequences selected from oligonucleotides and polynucleotides of viral RNA for the purpose of producing an RNA molecule containing those nucleotide sequences essential for self-replication or those nucleotide sequences essential for replication upon co-infection with complete TMV in a plant cell or cells, and having the potential, in the plant cell or cells, to express and control the expression of genetic information inserted thereby and those regions necessary for the production and accumulation of stable vector particles in the plant or plant cells. The production of a gene-derived product in a plant cell from the said at least a portion of an RNA vector is also described.

Modified seed storage proteins of maize (zeins) containing lysine and genetically modified structural genes encoding these proteins. The structural genes encoding the 19kd and 22kd zein proteins have been genetically modified so that the zeins are modified to contain lysine. These modifications are made without affecting the ability of the modified zeins to function correctly as a storage protein in maize endosperm. Various genetic constructs and vectors comprising these modified zein genes are provided.
