    摘要: The present invention relates to a novel biocontrol protein which is a crystal toxin and an insecticide and which is produced in a strain of bacteria genetically altered upon incorporation of a specific gene inserted as a stable gene construction. In particular, the present invention relates to the insertion of the δ-endotoxin gene from B. thuringiensis var. tenebrionis into a cloning vector containing prokaryotic regulatory and replication systems and the utilization of this vector to transform bacterial strains into coleopteran pathogens. Limited deletion of the coding region of the gene of this invention still allows preservation of the insecticidal activity in the protein expressed. In addition, this invention provides genetically altered bacterial strains that are insect toxic and that can be used as a novel method of biological control of agricultural insect pests.

    摘要: The promoter of the nitH gene of the fast-growing Rhizobium japonicum strain USDA 191, has been cloned. Over 4.2 kilobase pairs (kbp) of DNA were sequenced. Sequences encoding nitH and the 5'-end of nitD were identified, as were sequences involved in promoting operon transcription and a nifH ribosome binding site. Use of the nitH promoter to drive transcription in Rhizobium of heterologous structural genes is taught. Useful sequences and plasmids are also disclosed.

    摘要: Complete and partial Bacillus thuringiensis var. kurstaki HD-73 crystal protein protoxin genes were cloned into pRK290. E. coli cells harboring these recombinant plasmids produced protein that was toxic to Manduca sexta (tobacco hornworm) larvae. These plasmids were unstable in and inhibitory of the growth of Agrobacterium and Rhizobium cells; the presence of a complete protoxin gene being far more destabilizing and inhibitory than the presence of a partial protoxin gene. Rhizobium cells containing a partial protoxin gene formed root nodules in legume roots which were toxic to M. sexta larvae. Plasmids carrying complete protoxin genes were totally incompatable with root nodule formation; bacteroids in nodules formed by inoculation with stains containing the complete gene did not contain the gene-carrying plasmid and nodules were not toxic to insect larvae. The invention provides methods to produce insecticidal protein by constructing partial protoxin genes followed by expression of the genes in bacteroids and Rhizobiaceae cells. Useful plasmids and cells are also provided.
