    摘要: The present invention provides a method of enhanced protein production that comprises the step of expressing a recombinant gene encoding the protein in eukaryotic cells under conditions in which cleavage of the pro-domain of the protein is inhibited or eliminated. Generally the method of the present invention includes the step of inhibiting or altering the cleavage of a pro-domain of a recombinant protein of interest in order to increase the amount of recombinant protein secreted from a eukaryotic cell. Recombinant proteins that can be prepared using the method of this invention include members of the transforming growth factor-β (TGF-β) superfamily, such as bone morphogenetic proteins. Also provided are genetically engineered cells and polynucleotides for performing the method of the invention.

    摘要: A method of retaining growth factors on a substrate for release at a treatment region comprises immobilizing an antagonist of the growth factor on a substrate and binding of the growth factor to the antagonist. In one aspect, the substrate is provided on a bioimplant. The resulting bioimplant allows for activity of the growth factor to continue at the region of implantation. According to the method of the invention, exogenous growth factors can be used to stimulate the repair of various tissues and organs at the site requiring repair, and be protected from inactivation, sequestration or degradation. The invention also provides bioimplants and methods of delivering growth factors.

C., promotes osseointegration of an implant in bone, when introduced into a bore in the bone, prior to insertion of the implant in the bone; a method of placing an implant in bone comprises injecting said liquid composition into a bore in the bone prior to insertion of the implant in the bore, the TGF-.beta. being present in the liquid composition in a concentration effective in an interspace between an inner surface of the bore and an outer surface of the implant, to promote osseointegration.

    摘要: A bone mill comprising a vessel and a rotatable grinding tool to produce ground autologous bone particles for surgical bone reconstruction use, wherein the vessel includes a wall defining a circular cylindrical chamber having a concentric axis. The vessel further includes a bottom wall at right angle to the axis and a lid adapted to be removably connected to the vessel. The lid includes an opening coincident with the axis. The grinding tool includes an elongated shaft extending through the opening in the lid along the axis with a cutting blade mounted to the shaft within the chamber whereby the cutting blade can be rotated by motive device, such as a dental drill, engaging the shaft exterior of the vessel to cut bone, placed in the vessel, into said bone particles.
