A method for one-step immunoassay of human type IV collagen, which uses a carrier immobilizing monoclonal antibody against human type IV collagen and a labelled polyclonal antibody against human type IV collagen, as well as a kit therefor and monoclonal antibodies which are particularly suited therefor. It is possible to accurately and quickly measure human type IV collagen in blood serum, for example, and obtain measurement results which accurately reflect pathological conditions.

A type IV collagen high molecular form having a higher molecular weight than the 7S domain of type IV collagen and including the 7S domain in its structure, is obtained from the supernatant being recovered from a collagen solution digested by pepsin in the following steps;1) salt precipitating with sodium chloride at a concentration no higher than 1.2 M,2) dissolving the precipitates,3) salt precipitating with sodium chloride at a concentration no higher than previous concentration. By reacting a sample with an antibody which reacts with this form, the type IV collagen high molecular form in the sample can be measured to allow diagnosis of the degree of liver fibrosis in patients with liver diseases.

A sample containing hemoglobin A1c (HbA1c) is simultaneously brought in contact with a solid phase and anti-HbA1c antibody, the anti-HbA1c antibody bound to the adsorbed HbA1c on the solid phase and the anti-HbA1c antibody in solution are separated, and the anti-HbA1c antibody bound to the adsorbed HbA1c on the solid phase is detected. HbA1c % is determined by one-step immunoassay, and the time required for measurement is shortened in comparison with conventional immunoassay methods. Since the exposure time to the pretreatment solution present in the reaction solution during the immune reaction is shortened, formation of a precipitate in the sample as well as inactivation of the enzyme of the enzyme-labeled anti-HbA1c antibody, when used, is reduced.
