These methods may facilitate, e.g., the systematic reverse engineering, and optimization of mammalian cis-regulatory elements at high resolution and at a large scale. The method may include integration of multiplexed DNA synthesis and sequencing technologies to generate and quantify the transcriptional regulatory activity of e.g., thousands of arbitrary DNA sequences in parallel in cell-based as says (e.g., mammalian cell based assays).

摘要: Methods for alleviating the suppression of humoral immunity by myeloid-dependent suppressor cells (MDSCs) are provided which include the administration of a VISTA antagonist and an iNOS/NO inhibitor in a subject in need thereof, e.g., subjects with cancer or infectious disease wherein VISTA is aberrantly expressed. This combination has been discovered to elicit a synergistic or additive effect on alleviating suppression of humoral immunity by VISTA which is elicited by MDSCs.

摘要: Disclosed are Dpo4 DNA polymerase mutants with increased DNA synthesis processivity. The mutant enzymes, Dpo4 A181D and Dpo4 E63K, are constructed based on the wild-type Dpo4 derived from Sulfolobus solfataricus to obtain Dpo4DNA polymerase mutants with increased DNA processivity. The extension length of Dpo4 A181D and Dpo4 E63K are respectively increased by 25% and 18.8% than that of the wild-type Dpo4. The fidelity of Dpo4 A181D and wild-type Dpo4 are similar, and the fidelity of Dpo4 E63K is increased from that of the wild-type Dpo4. In summary, the mutants Dpo4 A181D and Dpo4 E63K obtained by the present invention have increased processivity compared to that of the wild-type Dpo4 DNA polymerase.

摘要: Embodiments of the present disclosure are directed to a heat exchange device that includes a condenser configured to receive a refrigerant, an evaporator having an evaporation tube bundle, a throttling device configured to receive a first portion of the refrigerant from the condenser and to expand the first portion of the refrigerant before directing the first portion to the evaporator, and an ejector having a high pressure conduit, a low pressure conduit, and an outlet conduit, the ejector is configured to receive the first portion from the throttling device or a second portion of the refrigerant from the condenser via the high pressure conduit, receive a third portion of the refrigerant from the evaporator via the low pressure conduit, mix the first portion or the second portion with the third portion to form a mixed refrigerant, and direct the mixed refrigerant to the evaporator via the outlet conduit.
