The present invention is related generally to embryonic-like stem cells isolated from adult human peripheral blood, designated herein as peripheral blood-stem cells (PB-SC), which display the characteristics of embryonic stem cells and hematopoietic cells. These cells have the capability of proliferation and are able to differentiate to other types of cells. These cells are, therefore, suitable for use in stem cell-based therapies, particularly autologous stem cell therapies, for the treatment of various diseases such as neurodegenerative diseases, autoimmune diseases, diabetes, spinal cord damage, multiple sclerosis, cardiovascular disease, stroke and birth defects.

The present invention relates to a purified population of fibroblast-like macrophage (f-macrophage, f-MΦ) and methods using the same. The f-MΦ can be expanded in vitro and differentiated into several lineages, including insulin-expressing cells, endothelial cells, and neuronal cells. The f-MΦ described herein have been generated from human umbilical cord blood (CB f-MΦ) and have characteristics similar to f-MΦ derived from peripheral blood. Thrombopoietin (TPO), at low dosage, significantly stimulates the proliferation of CB f-MΦ, wherein the TPO-expanded CB f-MΦ retain their pluripotent differentiation potential.

These cells have the capability of proliferation and are able to differentiate to multiple types of cells. In addition, the CB-SC display low immunogenicity and immune regulation. These cells are, therefore, suitable for use in stem cell-based therapies for the treatment of diseases such as Parkinson's disease, diabetes, spinal cord damage, multiple sclerosis, cardiovascular disease, stroke and birth defects, and for preventing, treating and/or reducing an autoimmune disease in a mammalian subject.
