    摘要: A virus-lysed tumor cell vaccine is an active immunotherapeutic agent against tumors in mammals. In particular, a vaccine based upon vaccinia virus-lysed, species-specific tumor cells is an effective stimulator of the immune response in mammals, and a vaccine based upon vaccinia virus-lysed spontaneously arising tumor cells is an effective stimulator of the immune response in some human cancer patients. A process for preparing the vaccine by viral oncolysis is also disclosed. Tumor cells are removed from a mammal, the cells are cultured in a culture medium and infected with live vaccinia virus. Viral oncolysis occurs during incubation and the resulting viral oncolysate, after extraction, may be injected into mammals to stimulate the immune response mechanism.

    摘要: A cell analyzer which automatically processes and evaluates cell samples for possible indications of malignancy and other abnormalities is disclosed. Cell samples are prepared in a suspension and are tagged with an appropriate radioactive stain or appropriate biochemical. Counting, filtering and drying steps under computer control are followed by the taking of a radioactive count with respect to a normalized number of cells. The results of the radioactive counting determine whether the particular sample is classified as "positive" or "negative".

    摘要: A Leighton tube assembly designed for the growth of tissue cultures and their preservation comprising a tube, a cover slip and a means to close said tube. The tube, made from two injection molded pieces welded together, has a chamber toward its closed end and a neck towards its open end. The chamber has a wall with a flat portion which is separated from the open end of the tube by a dam located in the neck. The cover slip is molded of a plastic material which is both transparent and inert with respect to most chemical solvents. The cover slip has a culture segment which lies on said flat portion of the chamber wall when the slip is in its growth position and a handle. The handle is of a length which allows the positioning of a cap over the open end of the tube.

    摘要: Continuous cell lines of macrophages are provided by the in vitro action, on mammalian macrophages withdrawn in situ, of macrophage activating substances, which cause the removal by the macrophages of the other cells present in the cellular medium withdrawn. The macrophages thus isolated are then placed in a suitable culture medium, until the establishment of a continuous line of macrophages, then the macrophages established as a continuous line are cultured in the presence of a suitable activating substance and the supernatant liquid is collected from these cultures. This supernatant contains substances inhibiting the proliferation of the cells having a rapid rate of multiplication. This supernatant liquid is, if necessary, subjected to a purification process. The product is notably useful for inhibiting human tumor cells.

    摘要: An apparatus and a method for preparing live, sterile cells from tissue are disclosed which enable high yields of viable isolated cells to be obtained with a greatly reduced possibility of contamination of the cells during the process. The apparatus and method allows one to truly mass manufacture isolated cells as contrasted to a small hand operation requiring many hours and dozens of people. The improved apparatus of the invention comprises a shredder device which mechanically shreds the tissue into small pieces of high surface to volume ratio, a processing vessel in which the shredded tissues are contacted with an enzyme solution to break down the intercellular material and thereby separate the cells one from the other and a means for separating the cells from the enzyme solution. Preferably, the means for separating the cells from the enzyme solution is a flow centrifuge which has been specifically designed to reduce shear force inside the bowl during operation. Also disclosed is a process for shredding tissue employing a plurality of moveable blade elements and at least a corresponding number of separated fixed elements. Interposing the tissue between the fixed and rotating elements and then driving the moveable blade elements through the spaces defined by adjacent fixed elements results in the tissue being shredded into small pieces of high surface to volume ratio. This in turn optimizes surface area for contact between the shredded tissue and the enzyme solution. In addition, a completely closed process is disclosed for obtaining cells from the tissue comprising the steps of sequentially shredding the tissue, contacting the shredded tissue at a controlled temperature with an enzyme solution to break down the intercellular material and then separating the isolated cells from the enzyme solution.

    摘要: Compounds capable of functioning as membrane mobility agents in plant and animal cells are described. These compounds have a structure represented by the formula ##STR1## in which A represents a hydrophilic chain; Z represents either an ester linkage ##STR2## an amide linkage ##STR3## or an ether linkage (--O--); R.sub.1 represents either a straight-chain alkyl group or a straight-chain alkyl group interrupted by one or more cyclopropyl groups; R.sub.2 and R.sub.3 each represent either hydrogen, lower alkyl, or a halogen group; and x represents an integer from 2 to 10.The membrane mobility agents of the invention promote the lateral mobility through cell membranes of molecules ranging in size from phospholipids to surface antigens. They represent a new class of biologically active molecules which may be used to alter the mobility of membrane components in animal and plant cells to alter such cells in their response to external stimuli. Among their uses are altering the immunological response of lymphocytes; altering the pharmacological response of target cells; altering the biological properties of cancer cells; and fusing two or more cells to create a new hybrid cell containing the genetic information from the fused cells.
